| Objective:To detect whether corneal lymphangiogenesis appears to follow with neovascularization in the alkali-burned rat corneal;the effect of corneal neovascularization and lymphangiogenesis after intervention with bevacizumab in the early period of alkali-burned rat corneal.Methods:50 sprague dawley,were randomly divided into two grous: Control group and Treatment group,each include 25 sprague dawley.50 sprague dawley were used to make corneal alkali-burned model by 1 mol/L NaOH.Control group was given 0.3%ofloxacin,Treatment group was given 0.3%ofloxacin and 4mg/ml bevacizumab.Different eye drops were applied twice daily for 14days.We observed corneal neovascularization on the 3d,5d,7d,14d after injury,after 5 rats of each group were randomly killed.Then examined the expression of VEGF and VEGFR-3 protein by immunohistochemistry on the 3rd,5th,7th,14th day after the injury respectively in the Control group and Treatment group.Then examined the expression of LYVE-1 protein by immunofluorescence on the 3rd,5th,7th, 14th day after the injury respectively in the Control group and Treatment group.In addition,the presence of blood and lymphatic vessels in the rat injuried corneas were examined with transmission electron microscope on 3rd,5th,7th,14th.Results:1,To observe the rats' cornea by slit lamp microscope:we found brush-angiogenesis at corneoscleral margin on the 3th day,new blood vessels invaded corneal on the 5th day,corneal neovascularization interlaced into mesh,some vessels penetrated alkali burn spot on the 7th day,corneal neovascularization almost occupied the entire cornea on the 14th day in control group.And brush-angiogenesis appeared some quadrant on 5th,corneal neovascularization invaded shallow layer of corneal stroma,however raritas and small on 7th day,corneal neovascularization distributed in the lateral of alkali burn spot on 14 th in treatment group.2,The expressions of VEGF,VEGFR-3 and LYVE-1 protein on the 3rd,5th,7th,14th day in Treatment group are significantly lower than Control group.The difference is significant(P<0.01).3,Blood vessels were identified on the 5th day and lymphatic vessels were identified in vascularized cornea on the 7th day by transmission electron microscope in contorl group.However,the time of neovascularization and lymphangiogenesis was delayed in Treatment group,the number of blood vessels and lymphatic vessels decreased significantly.Conclusion:1,Corneal lymphangiogenesis appears to follow with neovascularization in the early stage of alkali-burned rat corneal.2,Topically administered Bevacizumab(Avastin)at a concentration of 4mg/ml limits corneal neovascularization and corneal lymphangiogenesis in the early stage of alkali-burned rat corneal. |
