Objective: Gastric cancer is one of the most common malignant tumors in our country. The overall 5 years survival rate is less than 50%. The therapy includes operation,chemistry therapy,radiotherapy and combinated therapy now. But all these methods can not increase the rate of curing and surviving effectively, and the prognosis of most patients is worse. The gastric cancer is always diagnosed at the advanced stage, about 40%-50% of patients are in regional advanced stage. Modern medical experiments and clinical studies indicate that splenic asthenia has a close relationship with the development of gastric cancer, and the therapy may worsen the spleen asthenia which forms an infernal circle. In this experiment, we established a model of the gastric cancer subcutaneously transplanted in nude mice, and according to the administration, we tried to get the effects of Jianpi Yiqi Recipe on the growth of the tumors and the mechanism.Methods:1 Establishing of the model:BGC-803 cells were cultured in RPMI-1640 medium supplemented 10% new-born calf serum(NBCS) at 37℃, 5 %CO2. Logarithmically growing cells were used to establish a model as the first generation. And the experimental models were established by the sixth generation.2 Dividing groups:The nude mice were divided into four groups, negative control(NS) group, positive control(5-Fu) group, Jianpi Yiqi Recipe(JPYQR) group and combined(5-Fu+JPYQR) group.3 General state of nude mice:Monitoring the body weight, the drinking, eating and the mental state of the nude mice.4 Growth of tumors:Tumor volume was measured by vernier caliper, calculated according to the formula: Tumor volume(cm3) = ab2/2, where a, b are the major dimension, minor dimension. To draw the curve according to the tumor volume. Measuring the tumor weight and calculating the rate of tumor inhibition.5 Histological observation:The mice were sacrificed and the tumors were recovered for light microscope observation.6 Effects on immune function: Weigh the spleen; test the rate of NK cell.7 Flow Cytometry Assay:For DNA content and apoptotic rate analysis.Results:1 General state of nude mice:The mice in 5-Fu group were becoming depressed, drinking and eating less; other groups did not see this.Before the administration, there was no difference on body weight among the four groups(P=0.89). After the administration, compared with the NS-group, the body weight was decreased in 5-Fu-group(P=0.009) and combined-group(P=0.028); increased in JPYQR-group(P=0.295). Compared with the 5-Fu-group, the body weight was increased in JPYQR-group(P=0.047) and combined-group(P=0.754). Compared with the combined-group, the body weight was increased in JPYQR-group(P=0.076).2 Growth of tumors2.1 Tumor volume:Before the administration, there was no difference on tumor volume among the four groups(P=0.855). After the administration, compared with NS-group, the tumor volume was decreased in 5-Fu-group(P=0.019),combined-group(P=0.016) and JPYQR-group(P=0.675). Compared with the 5-Fu-group, the tumor volume was increased in JPYQR-group(P=0.047), decreased in combined-group(P=0.554). Compared with the combined-group, the tumor volume was increased in JPYQR-group(P=0.035).2.2 Tumor weight:Compared with the NS-group, the tumor weight was decreased in 5-Fu-group(P=0.012),combined-group(P=0.009) and JPYQR-group(P=0.461). Compared with the 5-Fu-group, the tumor weight was increased in JPYQR-group(P=0.173), decreased in combined-group(P=0.289). Compared with the combined-group, the tumor weight was increased in JPYQR-group(P=0.035). The tumor inhibition rate was 40.71%, 13.27% and 55.75% in 5-Fu-group, JPYQR-group and combined-group respectively.3 Histological observation:The cut surface was white in naked eyes; and with the light microscope, there were cellular necrosis aeras in all the four groups, and the necrosis aera is larger in 5-Fu-group and combined-group than NS-group and JPYQR-group.4 Effects on immune function4.1 Spleen weight:There was no difference on spleen weight among the four groups(P=0.863).4.2 Rate of NK cell:Compared with the NS-group, the rate of NK cell was decreased in 5-Fu-group(P=0.012), increased in JPYQR-group (P=0.009) and combined-group(P=0.295). Compared with the 5-Fu-group, the rate of NK cell was increased in JPYQR-group(P=0.047) and combined-group(P=0.35). Compared with the combined-group, the rate of NK cell was increased in JPYQR-group(P=0.035).5 Flow Cytometry Assay5.1 Analysis of cell cycle: For G2/M stage, there was no difference (P=0.248). For the G0/G1 and S stage, compared with NS-group, the number of tumor cell was increased in the combined-group(P=0.043). Compared with 5-Fu-group, the number of tumor cell was increased in the JPYQR-group(P=0.018) and the combined-group(P=0.039).5.2 Apoptotic rate:Compared with the NS-group, the Apoptotic rate was increased in the 5-Fu-group(P=0.031) and the combined-group(P=0.023). Compared with the 5-Fu-group, the Apoptotic rate was increased in combined-group(P=0.048).Conclusion:1 The JPYQR may have some effect on improving life quality after the therapy. For example: diet, spirit and general healthy state.2 The tumor inhibition ratio is 40.71%, 13.27% and 55.75% in 5-Fu-group, JPYQR-group and combined-group respectively. The JPYQR may have a synergic effect with chemotherapy.3 The JPYQR may be an effective composition for growth inhibition of gastric cancer and its mechanism may be related to strengthening the immune function and regulating the tumor cell cycle.
