The Research On Several Antihistamines Comparison Of The Anti-Inflammatory | | Posted on:2009-05-21 | Degree:Master | Type:Thesis | | Country:China | Candidate:R J Tan | Full Text:PDF | | GTID:2144360245488496 | Subject:Dermatology and Venereology | | Abstract/Summary: | PDF Full Text Request | | Objective: To observe the effect of some antihistamine drugs used frequently in clinic on leukotriene B4(LTB4),interleukin 4(IL-4) and interleukin 5(IL-5) released by spleen lymphocytes of allergic mice challenged by ovalbumin(OVA).Methods: Mice were sensitized by intraperitoneal injections of OVA on Days 1 and 15 to achieve allergic model of animals.The spleen lymphocytes were isolated and incubated with epinastine solution of different concentrations or other control solution prior to re-challenge with OVA. Double sandwich enzyme-linked immunosorbent assay(ELISA) was applied to detect the level of LTB4,IL-4 and IL-5 in cell culture supernates.Results: With the same concentration , The level of LTB4 in supernatant in epinastine group was significantly lower than that in ebastine group(P<0.01);the level of IL-4 in supernatant in epinastine group was significantly lower than that in cetirizine group(P<0.01);the level of IL-5 in supernatant in epinastine group was significantly lower than that in cetirizine and ebastine groups(P<0.01);With the same concentration,The level of LTB4 in supernatant in cetirizine group was significantly lower than that in ebastine group(P<0.05),The level of LTB4 in supernatant in cetirizine group was significantly higher than that in ebastine group(P<0.05)。Conclusion:With the same concentration,Epinastine,ebastine and cetirizine all express the anti-inflammatory effect more or less.
