The Correlation And Clinical Significance Of The Expression Of MCM2 And Survivin In HCC

Objective:To observe the expression of Survivin and MCM2 in the human hepatocellular carcinoma(HCC) and the correlation between them, investigate their effects on the occurrence, development and clinical pathological parameters of human hepatocellular carcinoma. To offer some laboratory foundations for the research of the molecular biological mechanism by which Survivin and MCM2 take part in the occurence and development of HCC.Method:From Jan 2003 to Dec 2005, 70 patients with HCC , which were treated in the first affiliated hospital of Nanchang University, were selected in this experiment with complete clinical datas, 10 normal liver specimens came from the lobectomy liver of patiens with intrahepatic bile duct stone. Those liver Specimens were studied by immemohistochemical Elivision^TM plus (AB) method to investigate the expression of Survivin and MCM2.The standard positive contract was from Ameican neomarkers biological company ; and the negative contract was PBS instead of Survivin,MCM2 as the first antibody in this experiment.SPSS14.0 for Windows statistical software was used to calculate the value of x² and Spearman's correlation coefficient.Results: It can be seen diffused distribution of buffy particles in the hepatoma carcinoma cell intracytoplasm by SP stain, these particles are Survivin protein. The positive rate is 65.7% and this number is 0 in control specimens, there is no significantly different between them. MCM2 protein diffused distributed in nucleus as buffy particles. The positive rate is 44.3% and this number is 0 in control specimens, there is no significantly different between them. Survivin protein which is expressed in HCC tissues have no significant correlation with gender, age, AFP, the size of tumor, the number of tumor, capsule infiltration and clinical stage, it is only related with whether there are portal vein cancer plugs or metastasis in liver(P< 0.05).But it can not observed significant correlations between MCM2 and above-mentioned clinical pathological parameters(P>0.05).Conclusions: 1. The high expression of Survivin and MCM2 correlated with the occurrence and development of human hepatocellular carcinoma;2. The high level of Survivin and MCM2 are not isolated events. There might be some synergistic actions between them in the regulation of the HCC occurrence; 3. The effects of Survivin and MCM2 in occurrence and development of human hepatocellular carcinoma offer some theoretical foundations for the research of clinical neotype molecule target pharmacal therapy.
