The Study The Effects Of Colchicine On Proliferation And Apoptosis Of Rat Glioma G422 Cells In Vitro, And Whether Colchicines May Damage The Sensorrimotor Functions

Objective:1.To study the effects of Colchicine on proliferation and apoptosis of rat glioma G422 cells in vitro.2.We injected colchicine into intracalvarium of rats and divided Into 4 treatmeant groups(64,32,16,8ug.ml-1)and observe the side effect of drug through ethology.Methods:1.The cells divided into many treatment groups(2,4,8,16 ug·ml-1 Colchicine and so on),and blank control group.The rat G422 cells were cultivated to 1×10~5/ml in the RPMI 1640 medium,then added 10ul into 96 holes board by 3 holes per group,and MTT assay was used to observe the proliferation of glioma G422 cells treated for 24,48and 72h. 2.We injected colchicine into intracalvarium of rats and divided Into 4 treatmeant groups (64,32,16,8ug.ml-1)and observe the side effect of drug through ethology.Results:1.MTT assay used to observe the proliferation of G422 glioma cells demonstrate that 4μg·ml-1,8mg·ml-1,16mg.ml-1 treated groups after 24h,48h,72h,suppressed G422 cell proliferation in a concentration and time-dependent manner indicated down-regulation than blank control group(P＜0.01).2.when the density is under 32ug.ml-1 the ethoiogy of rats are not change obviously.Conclusion:1.suppresse to G422 cell proliferation in a concentration and time-dependent manner indicated down-regulation than blank control group.2.when the density is under 32ug.ml-1 the ethoiogy of rats are not change obviously.
