The Expression Of CK19 In Sentinel Lymph Node Of Patient With Breast Cancer Detected By Loop-mediated Isothermal Amplification

Objective:To establish a more sensitive, objective and rapid assay to detect the metastasis of sentinel lymph node, helping the patient with breast cancer to improve the quality of life.Method:①Get the sentinel lymph node of patient with breast cancer by method of methylene blue, then detect the metastasis by traditional method of histopathology。②Detect the expression of CK19 in breast cancer and lymphoma cell lines by LAMP, to prove whether it can distinguish the cells derived from epithelium and lymph tissue。③Analysis the sentinel lymph node by LAMP, compare it's result to histopathology。Result:①successfully get 80 sentinel lymph node from 34 patient, 10 of them proved to be positive。②LAMP can distinctly distinguish breast cancer cells from lymphoma cells。③CK-19 gene can be amplified by LAMP from 10 positive SLN, the characteristic electrophoresis strip was showed, the result is completely accord with the histopathology,100% of coincidence.Conclusion:To detect the metastasis of sentinel lymph node by LAMP shows favourable application prospect, since it possess merits of specific, rapid and easy to manipulation. More importantly, it can detect the whole situation of the sentinel lymph node. In our later research procedure, we will try to quantitate the assay with fluorchrome, find the optimal reactive site, shorten the reaction time.
