Capillary electrophoresis was developed a powerful technique for seperation with the characteristic of high separation efficiency, rapid and small analyte consumption. Chemiluminescence analysis, based on the emission during some chemical reactions was a determination technique. It has been widely used in trace species analysis, biological engineering, molecule biology, medicine and environmental chemistry characterized with its high sensitivity,wide linear range and simplicity. Capillary electrophoresis coupled with chemiluminescence detection could meet the demand of high sensitivity and high separation efficiency for analysis of complex biological sample.Recently, nanoparticle has been received considerable interest in liquid-phase chemiluminescence research. It was reported that the different diament of gold nanoparticles could directly induce or catalyze Luminol-H2O2 liquid-phase chemiluminescence, and the drugs and biological active substances,which contain amino, hydroxyl, sulfhydryl and carboxyl, had vary degree to gold nanoparticles catalyzing luminol-hydrogen peroxide system by inhibition or enhancement. Based on this principle, the sensitive chemiluminescence method for detection of drugs or biological active substances can be established.In this work, the studies on drugs or biological active substances analysis by capillary electrophoresis coupled with gold nanoparticles catalytic luminol-H2O2 system chemiluminescence detection have been exhibited. With the sensitive capillary electrophoresis coupled with chemiluminescence instrument set up by ourselves, a new detection methods were developed to detect several drugs and biological active substances, and applied to detect the content of drugs in urine and serum. The main contents in this thesis consist of three parts as following:Part one: It has been found that gold nanoparticles (nano-Au) can enhance the chemiluminescence of the luminol-H2O2 system, but the epinephrine can inhibit these chemiluminescence signals in alkaline solution. Based on this inhibition function, a capillary electrophoresis coupled with chemiluminescence detection methed has been established for the determination of the epinephrine. Under the optimal conditions, the linear range for the determination of the epinephrine was 2.6×10-8~1.6×10-6 mol·L-1. with the detection limit of 6.9×10-9 mol·L-1 (S/N=3). This methed was applied to detect epinephrine in commercial epinephrine hydrochloride injection and in health human urine sample with satisfactory results.Part two: Based on the enhancement of chemiluminescence of luminol-H2O2 -gold nanoparticles system by norfloxacin, a novel and rapid capillary electrophoresis coupled with chemiluminescence detection method was established for determination of norfloxacin. The chemiluminescence intensity was proportional to the norfloxacin concentration in the range of 7.8×10-8~1.6×10-5 mol·L-1, with the detection limit of 7.3×10-9 mol·L-1(S/N= 3). The proposed method was applied to determinate the commercial norfloxacin tablets and study on pharmacokinetics of norfloxacin with satisfactory results.Part three: It has been found that gold nanoparticles (nano-Au) can enhance the chemiluminescence of the luminol-hydrogen peroxide system and the ascorbic acid can inhibit these chemiluminescence signals in alkaline solution. Based on the inhibition function, a capillary electrophoresis coupled with chemiluminescence detection methed has been established for determination of the ascorbic acid. Under the optimal conditions, the linear range for the determination of the ascorbic acid was 2.8×10-8~2.8×10-6 mol·L-1, with the detection limit 9.6×10-9 mol·L-1(S/N=3).This method was used for analysis of ascorbic acid in commercial vitamin C tablets and health human serum samples, and the results somewhat was consistent with that reported previously.
