| Objective.To approach the expression of PC cell-derived growth factor(PCDGF) during the rat lung squamous carcinogenesis.To analyze the correlation between PCDGF and squamous lung carcinogenesis.To study the value of the expression of PCDGF in lung cancer of Diagnosis and Staging.Methods:Seventy-two Wistar were randomly divided into control group(n=6),sham operation group(n=6),model group(n=60).Model group rats were instilled with 3-methylcholanthrene(MCA) and diethylinitrosamine(DEN)into the left lobar branchus to induce lung squamous cell carcinoma.To obtain specimen and serum in every pathological phase during the carcinogenesis,these rats were at different intervals.PCDGF monoclonalantibody was used for detection in the experiment,every pathological phase were observed under light microscope.The protein expressions of PC cell-derived growth factor in serum of during the lung squamous carcinogenesis process were examined by SA-ELISA method.Results:The specimens were measured by pathology,59 specimens of every pathological phase during the carcinogenesis showed:hyperplasia 10,squamous metaplasia 9, dysplasia 9,carcinoma in situ 11,infiltrating carcinoma 10 and metastasis 10.There are 12 normal lung sections in the normal group and sham operation group.(1).PCDGF was expressed in the serum of rats of normal control group,sham operation group and model group.(2).There was no significant difference in serum level of PCDGF between control group(0.1372±0.0088) and sham operation group(0.1380±0.0540)(P>0.05).(3).There was significant difference between in serum level of PCDGF control group(0.1372±0.0088) and modelgroups(0.2236±0.0329;0.2327±0.0545;0.2354±0.0337:0.2917±0.0600; 0.3010±0.0564:0.3045±0.0329)(P=0.000).(4).There was significant difference between in serum level of PCDGF sham operation group(0.1380±0.0540)(P>0.05)and model groups(0.2236±0.0329;0.2327±0.0545;0.2354±0.0337;0.2917±0.0600;0.3010±0.0564;0.3045±0.0329)(P=0.000).(5).The results of SA-ELISA indicated that PCDGF expression of metastasis(0.3045±0.0329) was significantly higher than other lung lesions (hyperplasia,squamous metaplasia,dysplasia,carcinoma in situ and infiltrating)(0.2236±0.0329;0.2327±0.0545;0.2354±0.0337;0.2917±0.0600;0.3010±0.0564)(P=0.000). PCDGF expression of hyperplasia(0.2236±0.0329) was no significantly differentwith the serum level of PCDGF in squamous metaplasia(0.2327±0.0545) and dysplasia(0.2354±0.0337)(P>0.05).There was significant difference between the serum level of PCDGF hyperplasia(0.2236±0.0329) with other pathological phases'(carcinoma in situ,infiltrating, metastasis)(0.2917±0.0600;0.3010±0.0564;0.3045±0.0329)(P=0.000).There was significant difference between the serum level of PCDGF in the lung squamous carcinoma in (0.2917±0.0600) and those of stages'(hyperplasia,squamous metaplasia,dysplasia) (0.2236±0.0329;0.2327±0.0545;0.2354±0.0337)(P=0.000).Conclusions:Our findings indicate that PCDGF may be play important roles in the lung squamous carcinogenesis.The intensity of PCDGF expression in hyperplasia and carcinoma in may indicate that it may be used as tumor marker to diagnosis the lung squamous squamous carcinoma in early stage. |
