Expression And Correlation Of E26 Transformation-specific 1 And Matrix Metalloproteinases 9 In Meningioma.

Objective : To study the expression of E26 transformation -specific 1(Ets-1)and matrix metalloproteinase9 (MMP-9) in meningioma and the correlation between Ets-1 and MMP-9. then to explore their role in the develpment and progression of meningiomaMethods : Collect 56 cases of meningioma and 9 cases of normal brain tissue. Divide 39 case of benign meningioma as benign group. 10 case of atypia meningioma and 7 case of malignant meningioma as malignant tendency group refering to 2000 world health organization nervous system tumor grading standard .9 cases of normal brain tissue as control group.Ets-1 and MMP-9 expression was detected by immunohistochemistry and RT-polymerase chain reaction (RT-PCR) in 3 group . then approach to the correlation between Ets-1 and MMP-9.Results : The immunohistochemistry results showed that Ets-1 was not or low expressed in control group, 15 case was positive to Ets-1 in benign group and 12 case was positive to Ets-1 in malignant tendency group. The positive rate of Ets-1 was significant higher in malignant tendency group compared with benign group(P < 0. 05). MMP-9 was not or low expressed in control group, 21 case was positive to MMP-9 in benign group and 14 case was positive to MMP-9 in malignant tendency group. The positive rate of MMP-9 was significant higher in malignant tendency group compared with benign group(P < 0. 05). Ets– 1 positive reaction was correlated with MMP-9(γ=0.284,P < 0. 05).Furthermore, the contents of Ets-1 mRNA in meningioma were significantly higher than they in control group(P < 0. 01).And they were also higher in malignant tendency group compared with benign group and control group(P < 0. 01). the contents of MMP-9 mRNA in meningioma were significantly higher than they in control group(P < 0. 01).And they were also higher in malignant tendency group compared with benign group and control group(P < 0. 01).Ets -1 mRNA level was correlated with MMP-9(γ=0.643,P < 0. 01).Conclusion :1.The expression of Ets-1 and MMP-9 are related to the pathological grades of meningioma, may be invoived in the tumorigenesis and progression of meningioma.2.The express of Ets-1 in meningioma is related to MMP-9. they may coordinate with each other in the procession of meningioma development. Ets-1 may regulate MMP-9 to the malignant procession of meningioma.3. The express of Ets-1 and MMP-9 in meningioma may be used as potential marker for the prognosis of meningioma.and may used as a new way in the therapy of tumor.
