Objective To Evaluate the Curative Effects and safety of temozolomide Combined with Radiotherapy in Treatment of Postoperative Malignant Glioma.Methods 55 postoperative patients with pathologically confirmed malignant glioma were randomly assigned to receive temozolomide concomitantly with radiotherapy(experiment group;n=21),125I intraradiotherapy(control group;n=15)and temozolomide alone(control group;n=19).The patients were followed up in long term for observation the time to progression,over all survival time and safety.Results Median time to progression was 56.33±3.36 weeks in experiment group,44.83±4.55 and 41.5±3.95 weeks in control group(P<0.05).Median overall survival time was 59.27±3.19 weeks in experiment group,50.19±4.80 and 47.65±3.97 weeks in control group(P<0.05).Only one patient experienced leukopenia and released with dose reduction treatment.Conclusion Temozolomide combined with radiotherapy seems to be more effective than intraradiotherapy or chemotherapy alone in malignant glioma.The combined treatment was well tolerated. Objective To establish quantitative real-time PCR method for detecting the expression of miR-125b in glioma and to investigate the relationship between miR-125b expression and tumor grades.Methods From 18 patients with different malignant glioma and 6 cases of normal brain tissue samples extract the total of microRNA,then through reverse transcription method obtain c-DNA,and then with fluorescent probe miR-125b fluorescence primer Quantitative PCR detected its CT value, using 2T-△△Crelative quantitative method gain the CT value of standardization, which compared miR-125b in the normal brain tissue and the expression of glioma.Results The expressions of miR-125b were 1.0732+0.1225,0.7914+0.06752,0.5392+0.0818,0.2497+0.05065 in normal brain tissue and gliomaⅡ,Ⅲ,Ⅳ,respectively(P<0.05).Conclusions The expression of miR-125b was lower in glioma than in normal brain tissues,and negatively correlated with glioma grade.This may be used for further study of miR-125b low expression mechanism, and tumor grades,malignancy and prognosis in glioma,achieving to utilize miR-125b in the diagnosis of gliomas and personalized treatment lay the foundation.
