| OBJECTIVE:To characterize the differential expression of high mobility group box-1 protein(HMGB1)mRNA in uterine leiomyoma and tissues 1 cm far away from the uterine leiomyoma,and to investigate the potential role of HMGB in pathogenesis and development of uterine leiomyoma.METHODS:The expression of HMGB1 gene in 42 cases of uterine leiomyoma and tissues 1 cm far away from the uterine leiomyoma were examined by real-time quantitative reverse transcription-polymerase chain reaction(real-time quantitative PCR).And the HMGB1 mRNA expression was compared in uterine leiomyoma and tissues 1 cm far away from the uterine leiomyoma.RESULTS:The expression of HMGB1 mRNA in uterine leiomyomas was higher than that in tissues 1 cm far away from the uterine leiomyomas,since the value of 2-ΔΔCTwas 12.38(>2).And the expression of HMGB1 mRNA in large leiomyomas uteri whose diameter were greater than 5 cm was higher than that in small leiomy0mas uteri whose diameter were 5 cm or less than 5 cm,the value of 2-ΔΔCTwas 3.86(>2). But the amount of HMGB1 mRNA in multiple uterine leiomyomas relative to that in single uterine leiomyomas was just 1.77(<2).CONCLUSIONS:HMGB1 mRNA showed up-regulation in uterine leiomyoma tissues,and so was in large leiomyomas than in small leiomyomas.It was suggested that HMGB1 mRNA may have some relationship with the growth and development of uterine leiomyoma. |
