BackgroundPulmonary tuberculosis is the communicable diseases caused by mycobacteria; the individual factors have an important influence on the process of tuberculosis. Up to now, many gene polymorphisms associated with TB were founded. The cell immunity is the main immune reaction after the invasion of mycobacteria, and macrophage is the important ingredient in cell immunity makes an important contribution to occurrence of tuberculosis.Both of NRAMP1 and Sp110 Gene have a functional correlation with macrophage. NRAMP1 protein can effect the epimembranal ion channel (Fe2+,Mn2+ ) of the phagosome, and the concentration change of the divalent cation will kill the bacteria or cellules in phagosome .Ipr1 Gene can regulate the mouse's hereditary immunity to tuberculosis. Sp110b protein in human has the highest homology with Ipr1 protein,Sp110 gene maybe a new candidate gene which has association with tuberculosis. Nowadays, the study report about Sp110 gene was very limitied, and the study were mainly taken in south African.The interaction between NRAMP1 Gene and Sp110 Gene need further investigation.ObjectiveTo study the association between NRAMP1rs17235409A/G,NRAMP1rs17235416TGTG/(-),Sp110rs3948464C/T polymorphisms and susceptibility to tuberculosis of the Han people in Chongqing, and analysis the interaction between NRAMP1 Gene and Sp110 Gene. This will be the foundation for revealing the cause of tuberculosis and preventing the tuberculosis.MethodUsing case-control study, we have identified 100 sputum smear positive TB patient from the tuberculosis hospital in Chongqing. A total of 106 Han people in Chonqing without history of tuberculosis and other disease were selected as control. Genotyping of the NRAMP1 rs17235409A/G,NRAMP1rs17235416TGTG/(-),Sp110 rs3948464C/T was done by PCR-RFLP. Using Chi-square test to analysis the association between the three sites and tuberculosis, the calculations and statistics were performed with SAS 8.1. Combination with test results doing in early days,the interaction analysis was done by the MDR software.Result1. The frequencies of NRAMP1rs17235409 GG,GA+AA genotypes in the case were 67.00%,33.00%, which were 87.34%,12.26% in the control, the difference between the case and the control group had statistically significance (χ2 =12.7567,P<0.05). NRAMP1rs17235409 GA+AA genotypes had a significant increased risk of tuberculosis (OR=3.5235,OR95%CI: 1.7427~7.0350) compared to GG genotype.The frequencies of NRAMP1rs17235416TGTG/(-) polymorphism had statistically significant difference between the case and the control group (χ2 =7.41, P<0.05), rs17235409 TGTG/(-)+TGTG(-)/(-) genotypes had a significant increased risk of tuberculosis (OR=2.3402, OR95%CI: 1.2688~4.3163) compared to TGTG/TGTG genotype.2. The frequencies of Sp110rs3948464 CC,CT genotypes had statistically significant difference between the case and the control group (χ2 =14.49, P<0.05). Sp110rs3948464 CC genotypes had a significant increased risk of tuberculosis (OR=4.3724, OR95%CI: 2.0453~9.3474) compared to CT genotype.3. There is no interaction between NRAMP1rs17235409A/G,Sp110rs722555A/G,Sp110rs11679983A/G and Sp110rs3948464C/T.Conclusion1. The NRAMP1 rs17235409A/G,NRAMP1 rs17235416TGTG/(-),Sp110 rs3948464C/T polymorphisms were possibly susceptible to tuberculosis of the Han people in Chongqing. The NRAMP1rs17235409A allele,NRAMP1rs17235416TGTG(-) allele and Sp110rs3948464C allele were possibly the risk factor of tuberculosis in the Han people in Chongqing district. 2. There is no interaction between NRAMP1rs17235409A/G,Sp110rs722555A/G,Sp110rs11679983A/G and Sp110rs 3948464C/T.It is the first time to study the relationship between Sp110 rs3948464C/T and tuberculosis in China,and it is also the first time to study the relationship between NRAMP1 gene and tuberculosis in Chongqing district. All the results will help to explain the pathogenesy of TB, protect the high-risk group and to develop the medicine.
