Study Of The NRAMP1 Gene Polymorphisms With Susceptibility To Tuberculosis In Han Population In Qinghai

Objective:To study the relationship between NRAMP1 gene polymorphisms and susceptibility to tuberculosis in Han population in Qinghai,and to provide some basic research for the genetic aspects of pulmonary tuberculosis in Qinghai province,and to provide some theoretical basis for future precision medical treatment.Method: A case-control study was conducted in this study.Collected peripheral blood from 102 Han patients with pulmonary tuberculosis and 104 healthy subjects in Qinghai province.The two loci of rs17235409(D543N)and rs3731865(INT4)were genotyped by real-time fluorescence quantitative PCR-Taq Man probe method.The genotyping of rs17235416(3'UTR)was detected by PCR-RFLP.To analyze the association between the polymorphism of 3 sites and tuberculosis.Results:The genotype frequencies of rs17235409 G/G and G/A + A/A polymorphism were 74.5%?25.5% in the case group,and 80.8%?19.2% in the control group.There was no significant difference in the genotype frequencies between the two groups(X2=1.163,P=0.281).The genotype frequencies of rs3731865 G/G and G/C + C/C polymorphism were 72.5%?27.5% in cases,and 72.1%?27.9% in controls.No significant difference in the genotype frequencies between the two groups(X2=0.005,P=0.945).The genotype frequencies of rs17235416 TGTG/TGTG and TGTG/del + del/del were 74.5%,25.5% in patients,and 87.5%,12.5% in controls.There was significant differences in the genotype frequencies between the two groups(X2=5.662,P=0.017),OR(95%CI):2.395(1.152-4.980).The TGTG and del alleles in the case group were statistically significant compared with the control group(X2=9.878,P =0.002),OR(95%CI): 2.771(1.439-5.338).Conclusion:The rs17235409 and rs3731865 polymorphisms of NRAMP1 gene were not associated with susceptibility to tuberculosis in Han population in Qinghai.However,the NRAMP1 gene polymorphisms at rs17235416 site was involved in the susceptibility to tuberculosis in Han population of Qinghai.The rs17235416 TGTG/(del)was a risk factor for tuberculosis in Han population in Qinghai.
