| Objective: The main aim of this study was to investigate the changes about Podocytes ultrastructure,Proteinuria and the expression of Nephrin ,Podocin and CD2AP in diabetic nephropathy(DN) rats,and the relationship among these changes,observe the effects of candesartan influence on Podocyte.Methods: 36 Sprague-Dawley rats were divided at random into normal control group (A , n=12),DN model group(B , n=12),treatment group with candesartan(C ,5mg/Kg.d n=12). 24h urinary Protein count , blood glucose level , body weight and Scr et al were measured after 4 weeks of treatment, Pathological changes were evaluated by HE stain in renal tissues; and the ultrastructures of Podocyte were observed by electron microscope,and the expressions of nephrin,podocin and CD2AP Were detected by RT-PCR.Results: group B compared with group Aâ‘ 2 4h urinary protein count,Scr increased and Ccr decrease in group B (p<0.05).â‘¡The expressions of nephrin,Podocin and CD2AP's mRNA reduced in group B (P<0.05).â‘¢The foot process width (FPW) and thickness of GBM increased in group B (p<0.01). After treatment with candesartan:â‘ The levels of 24h urinary protein count, Scr all decreased and creatine clearance ratio (Ccr) increased comparing with group B (p<0.05).â‘¡The expressions of nephrin, Podocin and CD2AP's mRNA increased (P<0.05)â‘¢FPW and GBM decreased comparing with group B (p<0.05).Conclusion: Podocyte ultrasturcture and the expression of nephrin, podocin and CD2AP have changed in DN. The mechanism of candesartan which reduced urinary Protein count may act through up-regulating the expressions of nephrin, Podocin and CD2AP. |
