| Artificial oxygen carriers are products that can expand blood volume, maintainosmotic pressure and deliver oxygen to tissues, temporarily substituting for the humanblood. For the reasons of HIV epidemic, natural disasters, wars, et al, research onartificial oxygen carrier has attracted much attention during recent years, and muchprogress has been made. Polymerizing hemoglobin with cross-linkers likeglutaraldehyde can lengthen the hemoglobin's half-life in vivo and reduce the kidneytoxicity of bovine hemoglobin, as well as the immunogenicity. Moreover, theabundance of bovine blood has made research in this area a hotspot.The whole project is composed of two relating parts:First is the extraction and purification of bovine hemoglobin using centrifugationand heating treatment. After several repeats, the mean yield of the bovine hemoglobinhas reached 80%, with the purity of 96% or so.Second is the polymerization of bovine hemoglobin with glycolaldehyde. Everyproduct of a different condition was examined carefully by SDS-PAGE, in the aim ofoptimizing the reaction.The only goal for the project is to set up a whole routine, composed of thepurification of hemoglobin, its consequent polymerization, the systemic analysis ofproduct, and even the final clinical test, for the development of hemoglobin basedoxygen carreiers.Even though I didn't finish all the job, still, my hard working has built a stongfoundation for the following research. |
