Background: Chemotherapy is one of the common cancer treatment methods,but the non-specific killing of normal cells by chemotherapy drugs can cause different toxicity and side effects.Bone marrow suppression is one of the common toxic side effects.When bone marrow function impaired,white blood cells and red blood cells will decrease,leading to low immunity,infection,anemia.Cyclophosphamide(CTX)is one of the representative chemotherapeutic drugs.Its metabolites not only inhibit the division of tumor cells,but also kill bone marrow cells with high division activity.Cyclophosphamide can also cause liver and kidney damage.The current treatment of myelosuppression cannot avoid the dose reduction of chemotherapy drugs after the onset of myelosuppression symptoms,and may reduce the anticancer efficacy.Our Previous study shown that glutaraldehyde polyhemoglobin(pPolyHb)has a therapeutic effect on anemia rats caused by chronic kidney disease.Therefore,in this study,bone marrow suppression induced by cyclophosphamide in mice to explore the therapeutic effect of pPolyHb.Methods: The modeling scheme was verified by body weight and peripheral blood images of mice.From day 1 to day 10 of the experiment,mice were intraperitoneally injected with cyclophosphamide 25 mg/kg daily to induce bone marrow suppression.The subsequent treatment was continuous intraperitoneal injection of pPolyHb into mice during and after model building.On days 6,11,13,14,16,and 18 of the experiment,the peripheral blood images were measured by hematology analyzer to evaluate the status of bone marrow suppression and its changes.The index of spleen and thymus and swelling rate of the right ear were calculated.Serum IFN-γ and IL-4 concentrations were measured to analyze the immune promotion effect of pPolyHb.The index of liver and kidney was calculated and the concentrations of glutamic-pyruvate transaminase,glutamic-oxalacetic transaminase,creatinine and urea in serum of mice were measured by blood biochemical analyzer to explore the protective effect of pPolyHb on liver and kidney.Results:(1)The peripheral blood images of mice injected intraperitoneally with 25 mg/kg cyclophosphamide for 10 days showed a significant decrease in WBC and RBC.Bone marrow suppression was successfully established.(2)50 mg/kg pPolyHb significantly increased erythrocyte and hemoglobin levels in bone marrow suppression mice,and(3)50mg/kg pPolyHb decreased liver index,alanine aminotransferase(ALT)and aspartate aminotransferase(AST)on day 13 of the experiment.(4)The combined treatment of 50mg/kg pPolyHb and 10 mg/kg p Hb can not only improve erythrocytes,but also regulate the immune function in myelosuppressive mice.Conclusion: This study showed that 50 mg/kg pPolyHb had therapeutic effect on bone marrow suppression induced by cyclophosphamide,and a protective effect on liver,and when it combined with 10 mg/kg p Hb,the therapeutic was the best.
