Cardiovascular disease is one of the most fateful diseases in the world, and atherosclerosis is the most important foundation in the pathology of Cardiovascular disease. Numerous epidemiologic studies have shown that major risk factors of CVD were hypertension, dyslipidemias, high-level low density lipoprotein, cigarette smoking, diabetes mellitus, obesity, and most of them are associated with atherosclerosis. In recent years, many researches have indicated that initial factor and developing process of atherosclerosis and its complications seem to be like the process of inflammation; therefore, they have believed that atherosclerosis is a kind of special and chronic inflammation process. Local and systemic inflammation may play an important role in the initiation and progression of atherosclerosis and its complications. Up to now, the importance of correlation between CVD and inflammation has been increasingly emphasized. Periodontal infection is the most common infectious diseases in human beings. Besides the direct destruction caused by pathogens, the most major reason is that the host immune reaction to inflammation may cause periodontal tissue destruction. Epidemiological researches have shown that there has been close associations between periodontitis and the CVD and chronic periodontitis may be an independent risk factor for CVD. The aim of this study is to establish a new animal model of periodontal infection accompany with atherosclerosis in minipigs. Immunohistochemical stain and histological observation were carried to evaluate the atherosclerotic pathological changes in the aorta of animal model, and the concentration of some cytokines were detected. The results can provide evidence of the correlation between CVD and periodontal disease.Method: four 7-month-old WUZHISHAN minipigs were divided into three groups to establish a new animal model of periodontal infection accompany with atherosclerosis by ligation, wound, inoculated with Porphysomannas gingivalis and fed high fat diet. The periodontal indices, the serum lipoprotein levels and the concentration of CRP, IL-18 were examined at different intervals.Result: The minipig models have been established successfully.The compound groups showed deeper periodontal pockets and higher score of sulcus bleeding index(SBI) than other groups, the serum lipoprotein levels of TC, LDL-C, HDL-C ,CRP and IL-18 were increased in compound groups.Conclusion: The more severe atherosclerotic pathological changes can be found in the aorta of the compound animal models than in the control models. With the help of stimulation of periodontal pathogens, and the immune response, periodontal infection may play an important role in the development of atherosclerosis, but the potential biological mechanisms should be studied further.
