| Objective:To study the effect of Coumarins of Fructus Cnidium(TCFC)on osteoporosis (OP)in ovariectomized rats,investigat its mechanism and provide experimental theoretical basis for clinic.Methods:Sixty female Wistar rats were randomly divided into six groups(n=10):The sham operation group,ovariectomized group,Xianlinggubao capsule group(270mg/Kg), low dosage TCFC(10mg/Kg)group,middle dosage TCFC(20mg/Kg)group,high dosage TCFC(40 mg/Kg) group.Besides sham group,the other rats in each group were performed ovariectomy on both sides.Drugs were given from 24hours after surgery,once a day,12 weeks in total.After the 24 hours of last administration,rats were taken out blood in abdominal aortic and narcosis by 3%Chloral hydrate aldehyde(10ml/Kg).The level of estradiol(E2),calcium(Ca2+),phosphorus(P)and alkaline phosphatase(ALP) activity in serum were measured according to Kit;The tibia was taken out to determine the level of Ca2+,P magnesium(Mg),natrium(Na),The femur were taken out to determine the bone mineral density(BMD),hiomechanics properties and morphometry.Results:(1) TCFC compared with the ovariectomized group,ALP activity in serum were significantly decreased in low dosage group,middle dosage group,high dosage group(P<0.05,P<0.01,P<0.01)。(2) TCFC compared with the ovariectomized group,distance of bone trabecula of rats were significantly decreased in middle dosage group and high dosage group(P<0.05,P<0.01)。(3) TCFC compared with the ovariectomized group,the level of E2 in serum were significantly increased in middle dosage group and high dosage group(P<0.05,P<0.01)。(4) TCFC compared with the ovariectomized group,the level of Ca2+ in serum of rats were significantly increased in high dosage group(P<0.05)。(5) TCFC compared with the ovariectomized group,the level of Ca2+in tibia of rats were significantly increased in middle dosage group and high dosage group(P<0.05,P<0.01)。 (6) TCFC compared with the ovariectomized group,the level of Mg in tibia of rats were significantly increased in middle dosage group and high dosage group(P<0.05,P<0.05)。(7) TCFC compared with the ovariectomized group,BMD of the femur metaphysic of rats were significantly increased in high dosage group(P<0.05)。(8) TCFC compared with the ovariectomized group,thickness of cortical of rats were significantly increased in middle dosage group and high dosage group(P<0.05, P<0.01)。(9) TCFC compared with the ovariectomized group,maximal load of femur of femur were significantly increased in low dosage group,middle dosage group and high dosage group(P<0.05,P<0.01,P<0.01)。(10) TCFC compared with the ovariectomized group,ancyl-elastic modulus of femur of femur were significantly increased in low dosage group,middle dosage group and high dosage group(P<0.01,P<0.01,P<0.01)。(11) TCFC compared with the ovariectomized group,anti-cyrto-intensity of femur of femur were significantly increased in low dosage group,middle dosage group and high dosage group(P<0.05,P<0.01,P<0.01)。(12) TCFC compared with the ovariectomized group,the number of trabecular of femur were significantly increased in middle dosage group and high dosage group(P<0.05,P<0.01)。(13) TCFC compared with the ovariectomized group,percentage of trabecula of femur were significantly increased in middle dosage group and high dosage group(P<0.01,P<0.01)。(14) TCFC compared with the ovariectomized group,width of trabecular of femur were significantly increased in high dosage group(P<0.05)。Conclusions:TCFC had antiosteoporotic effects on ovariecmized rats.The mechanism has relationship with its raising the level of E2 in serum and reducing of ALP activity to depress the hyper bone turnover. |
