OBJECTIVE:To reveal the mechanism of acupuncture on ischemic brain injury, the article studied the effects on transportion function of microvascular endothelium cell, the expression of associated protein and the hippocampus perfusion of middle cerebral artery occlusion(MCAO) rats treated with acupuncture on Neiguan.METHOD:The model was established on rats by right MCAO induced by thread ligature. Rats was randomly divided into four groups:sham operation group, model group, Diji group, Neiguan group. The last two groups were freated with electro-acupuncture for 30 minutes per day. The change of perfusion in hippocampal CA1 area was monitored by Laser-Doppler Flow meter(LFD) on the day of 1, 5, 10days after the model was acupunctured. St aining with HE, the morphologic change of hippocampal neuron was closely observed, quantitative analysis of apoptosis cells was carried out with Image Pro Plus vesion 4.5. The contents of glucose, lactic acid, pyruvic acid,and Ca2+, Glutamic Acid in brain tissue were analyzed with different measuring image processing software kits respectively. The expression of Glut-1, MDR-1, HIF-1αwas observed with immuno- histochemisty assay.RESULTS:Electro-acupuncture on Neiguan can improve blood flow in hippocampal CA1 area, and decrease the death rate of neurons in hippocampal CA1 area, and shoeing a tendency of apoptosis inhibition. Electro-acupuncture on Neiguan can also increase the contents of glucose, pyruvic acid in brain tissue, and reduce the contents of lactic acid, Ca2+, Glutamic Acid. Electro-acupuncture on Neiguan can increase the expression of HIF-1αin hippocampal CA1 area in MCAO model group; There was a trend to increase the expression of Glut-1,P-gp, but with no statistic mean.Conclusion:Electro-acupuncture can improve blood flow in hippocampal CA1 area, enchance the transporting function of microvascular endothelium cell in rats with MCAO. Which was explain the relieves of the neuroal damage after acapnnctane in rats with MCAO.
