| Objectives:The effects of acupuncture on the transporting function of microcascular endothelium cell after ivc 5-thio-d-glucose and cyclosporin A. For the acupuncture and provide experimental and theoretical basis treatment of cerebral ischemia disease with acupuncture.Methods:Wistar rat suture method was induced with thread sature wistar rat focal cerebral ischemia model produced (MCAO), of the middle cerebral artery occlusion Intracerebroventricular microinjection of Glut-1 and P-gp blocker was applied then. Electric acupuncture on weiguan was given to the rats with electic stimulator, using the rat,s pial microcirculation.,was mouitored with laser Doppler flowmetry the content of on the 7th day Glutamate, lactate, pyruvate, Ca+ in cortex, hippocampus, thalamus and other brain regions wear measured the morphological changes of hippocampal neurons wea observed in HE staining slide; the use of of the Glut-1, MDR1-encoded P-gp in microvascular endothelial cell was studied with immunohistochemistry methed. hippocampal CA1 areaResults:Acupuncture can improve in microcirculation blood flow quantity in MCAO rats. Acupuncture on neiguan can reduce brain Glutamate, lactic acid, Ca+, increased pyruvate and increace serumglucose and glucose in cortex, hippocampus, thalamus and other brain regins. Acupuncture in neiguan can increase the number capillaries in hippocampal CA1 area, promote neuronal repair.It is showed that acupuncture can improve the expression of Glut-1 in microvascular endothelial cell hippocampal CA1 area; MDR1 encoded P-gp expression in the acupuncture it also significantly increased.Conclusions:Acupuncture can improve the microcirculation blood flow in MCAO rats, promite the expression of Glut-1 and MDR1-encoded P-gp in microvascular endothelial cell and reduce neuronal damage after ischemia. Acupuncture can improve the transport function of endothelial cells. |
