| The occurrence and development of lung cancer are closely related with dysfunction of the immune against tumor and immune escape of the tumor,and the creation of Foxp3+CD4+CD25+ regulatory T cells means an important mechanism, which makes the tumor specific and effective T cells can not expand to such a level as to eradicate the tumor. Recent study showed that forkhead / wingedhelix transcription factor( Foxp3 ) expressed especially on Treg cells,and was greatly related with its maturation of development and function.Objective To analyze the relation of Foxp3+CD4+CD25+ regulatory T cells and different histological types,various pathological stages,lymph nodes metastasis,surgery operation of the patients with lung cancer, investigate its role in tumor immunoregulation and its clinical significance.Methods1. Separation of mononuclear cells of peripheral blood(PBMC) and tumor infiltrating lymphocytes of tumor tissues (TIL) Density gradient centrifugation was used to isolate the mononuclear cells from peripheral blood of 14 normal controls and 44 patients with lung cancer of pre-and-post operation;The fresh tumor tissues were collected during operation and the tumor infiltrating lymphocytes were extracted afterwards.2. RT-PCR was performed to to analyze the expression of Foxp3 mRNA in peripheral blood and tumor tissues of the patients with lung cancer and the normal controls.Results1. The expression of Foxp3 mRNA in tumor tissues was higher significantly than that in preipheral blood of patients with lung cancer and normal controls,the same result was found in peripheral blood between the patient and normal control.2. The expression of Foxp3 mRNA was related to the size and local invasion of tumor ;Among different histological types, Foxp3 mRNA showed a highest expression in cases of small cell lung cancer,the second was adenocarcinoma and adenosqumous carcinoma, and the lowest was squamous cell carcinoma;it was also significantly correlated with tumor TNM stages and lymph nodes metastasis,the later TNM stage or lower tissue differentiation,positive lymphnode metastasis case showed a higher expression.3. The expression of Foxp3 mRNA decreased notably post-operation.Conclusion1. Foxp3+CD4+CD25+ regulatory T cell-mideated immune suppression is related to the occurrence, development and prognosis of lung cancer, surgical resection of tumor may reduce the bearing-tumor environment and Treg cells,so as to release the immune suppression of tumor,and will be helpful to the recovery of immune function on postoperative patients.2. To select effective biological therapy,down-regulate the expression of Foxp3 gene and decrease the quantity and function of Foxp3+CD4+CD25+ regulatory T cells will help to improve anti-tumor immunity of the body and the treatment level on lung cancer. |
