| ObjectiveColorectal cancer(CRC) is a common malignant tumor,which morbility keeps rising in China.Screening of colorectal cancer is a public health priority.Mass screening for colorectal cancer can reduce mortality.The most extensively screening method of CRC, faecal occult blood testing(FOBT),will always be limited in sensitivity.Our experiment is conducted to study and evaluate the expressions of human heat shock Protein 60(hHSP60) of colorectal cancer in human serum and faeces.Materials and Methods1.Selection and group of patients:40 patients with CRC were treated with radical resection in Department of General Surgery,Research Institute of Surgery,Daping Hospital,Third Military Medical University from October 2006 to January 2007.Among these 40 patients,28 cases were male,12 cases female,mean age 57 years old;22 cases were suffered from rectal cancer,18 cases from colon cancer;38 cases were with adencarcinoma, one case with neuroendocrine carcinoma,one case with mucinous adenocarcinoma.2.Methods:The hHSP60 was detected by using ELISA method in human serum and faeces from normal controls and pathologically proved patients before operation,and the correlation of their expressions with clinic-pathological variables were evaluated,hHSP60 was stained immunohistochemistry and detected by western blot in 40 CRC tissue and 20 corresponding normal tissue.Reuslts1.Expression of hHSP60 in CRC tissue was higher than normal colorectal mucosa.2.Compared with normal control,hHSP60 of cases with colorectal cancer was increased significantly in human serum(p=0.032),no associated with tumor location,gross type,histological differentiation,lymph node involvement,invasion depth and TNM stage. 3.Compared with normal control,hHSP60 of cases with colorectal cancer was increased significantly in human faeces(p=0.001),which was higher than in serum(p=0.000). hHSP60 in stool of CRC was related to gross type(p=0.042),invasion depth(p=0.028) and metastasis(p=0.045).No significant difference were found between hHSP60 in stool of CRC and tumor location,lymph node involvement,histological differentiation and TNM stage.4.The sensitivity of hHSP60 test in human serum for identifying CRC was 35%with specificity of 72.2%.hHSP60 test in human faeces identified 55%of CRC with specificity of 79.9%.Conclusions:1.Expression of hHSP60 in CRC tissue was higher than normal colorectal mucosa.2.hHSP60 content of cases with CRC was increased significantly in human serum.3.hHSP60 content of cases with CRC was increased significantly in human faeces,and higher than in human serum.hHSP60 content in faeces of CRC with polypoid/large fungating tumor was increasing significantly.4.Detecting of hHSP60 in faeces will be a method of screening CRC. |
