| The study was designed to extract Artemisia argyi essential oil sufficiently,select suitable dosage form,and investigate the effect of Artemisia argyi essential oil on 12 common pathogenic bacteria,we would evalute the immune function by whole level, cellular level and molocular level,the results are reported as follow:(1) The essential oil was etracted from Artemisia argyi by steam distillation,and Prepared stable emulsion by traditional emulsiyfing technique with dissolvent (Tween80).(2)The antibacterial effect of Artemisia argyi essential oil was measured by the Kirby-bauer(K-B),its minimal inhibitory concentration(MIC) and minimal bactericidal concentration(MBC) were by double dilution tests.In the experiment of antibacterial activity against Escherichia coli,staphylococcus aurcus,anthrax Bacillus in livestock and poultry;Escherichia coli,pseudomonas fluorescens,Aeromonas hydrophila,Klebsiella oxytoca,Myxococcus piscicola,vibrio parahaemolyticus,the MIC of which are 1×10-4mL/mL,1×10-4mL/mL,0.063×10-4mL/mL,1×10-4mL/mL, 0.5×10-4mL/mL,0.5×10-4mL/mL,2×10-4mL/mL,2×10-4mL/mL,1×10-4mL/mL.The MBC of Artemisia argyi essential oil to Escherichia coli,staphylococcus aurcus, anthrax Bacillus in livestock and poultry;pseudomonas fluorescens,Aeromonas hydrophila,Myxococcus piscicola are 1×10-4mL/mL,1×10-4mL/mL,2×10-4mL/mL, 0.5×10-4mL/mL,0.5×10-4mL/mL,2×10-4mL/mL.Artemisia argyi essential oil has obvious antibacterial activity.(3) Blood cell was tested by Hematology Instrument,blood biochemistry indexs were tested by kit.the result showed:compared with the control group,Artemisia argyi essential oil has no obvious influence on the Blood cell,There were significant difference in TP,ALB,SOD,AKP,LZM level.Especially the levels of AKP in three groups were extremely increased.It showed Artemisia argyi essential oil can improve the nutritional status of the body and improve immune function.(4) The appreciation ability of spleen lymphocyte,carbon clearance index,organ index,delayed type hypersensitive(DTH)and serum hemolytic antibody were selected to observe the effects of Artemisia argyi essential oil on the immune function by conventional methods.Compared with the control group,there are all significant difference upwards except for spleen index.It showed that Artemisia argyi essential oil can improve immunue function.(5) The mRNA expression level of IL-2,IL-4,IFN-γ,in speen of mice were examined by RT-PCR.The results showed:Compared with the control group,the mRNA expression of IL-2 in three groups increased.mRNA expressions of IL-2 in splenocytes could be prorated in both the M and the H group(P<0.01).The levels of IL-4 mRNA expression in both the M group and the H group were extremely increased(P<0.05).However No significant difference in the levels of IFN-γ,mRNA expression,certain dose of Artemisia argyi essential oil can improve the mRNA expression level of IL-2,IL-4 expression,thereby enhancing immune function and resistance to disease.To sum up,The method of extraction is correct,simple,the emulsion of Artemisia argyi essential oil has good stability.The results show that:Artemisia argyi essential oil has obvious antibacterial activity and improves immune function by whole level,cellular level and molecular level.
