| As a kind of Chinese herbal medicine,Artemisia argyi is not only medicinal but also edible.It is also a broad-spectrum antimicrobial and antiviral drug.It can inhibit and kill many viruses and bacteria,and has a certain preventive and therapeutic effect on respiratory diseases.Although the value of Artemisia argyi has been well known and many products of Artemisia argyi have been developed,there is no systematic and relevant report on whether the extract of Artemisia argyi can be used as healthy products and killing microbial medicine.In order to study in detail whether the extract of Artemisia argyi can be used in daily health care and beriberi treatment,we carried out this research.Firstly,several microorganisms are collected and processed.Plates carrying sealed solid medium for cultivating bacteria and fungiwere used to collect bacteria and fungi air by air deposition at different collection locations in Nanyang City(Nanyang Normal University,Nanyang Jiangying Airport,Nanyang Stadium,and Xinhua City Square),then brought them back to the laboratory for cultivation and isolation of related strains.At the same time,the bacteria and fungi associated with beriberi were screened and cultured by sterilized double steaming water immersion method.Twenty-eight strains were selected from about 60 plates of culture medium for microbial classification identification.The conservative sequences of 16 S and ITS of these strains were amplified by PCR and sequenced.By BLAST analysis,18 strains of bacteria and 10 strains of fungi were found,of which bacteria including Bacillus accounted for 90%.Fungi include Penicillium oxalate,Aspergillus Echinococcus and Alternaria,among which F-HZJ1,F-HZJ2 and F-ZBJ2 are Penicillium oxalate.According to the characteristics and frequency of various strains,especially harmful strains must were selected.Ten strains were selected from these identified strains,including 6 strains,B-JQ(Acinetobacter baumannii),B-SY(Bacillus megacephala),B-SY7(Stenotrophomonas rhizophilus),B-JC1(Bacillus subtilis),B-XH1(Bacillus amylolytica)and B-SYS2(Escherichia coli).Four fungi were F-HZJ2(Penicillium oxalate),F-SY1(Alternaria tenuifolia),F-SYS1(Penicillium expansum)and F-JC3(Mycospora mycospora).Secondly,the antimicrobial activity of the extracts of Artemisia argyi obtained by deionized water extraction and Soxhlet extraction and commercialized CEA extracts of Artemisia argyi to 10 tested strains was tested.The phenotype observation of the antimicrobial effect of the extracts of Artemisia argyi confirmed that the extracts of Artemisia argyi could inhibit these microorganisms.Finaly,through detailed physiological measurement and data statistics,quantitative determination and data analysis of the inhibitory effects of extracts from Artemisia argyi on the growth of several isolated microorganisms were carried out to confirm the efficacy of extracts from Artemisia argyi.It was found that different extraction methods and treatment methods had different inhibitory effects on bacteria.The three extracts had different inhibitory effects.BEA had obvious inhibitory effect on bacteria from the surface,but almost no effect on fungi.The extracts of SEA and CEA had certain inhibitory effects on the 10 tested bacteria.The inhibitory effects of SEA and CEA on four fungi were stronger than that of six bacteria.Different concentrations of SEA and CEA have different inhibitory effects on different strains,but overall the inhibitory effect of SEA extracts on bacteria is better than that of CEA.SEA and CEA extracts have better inhibitory effect on Acinetobacter baumannii and Penicillium oxalate.Essentially,the concentration of SEA and CEA extracts is 25 mg/ml for all tested bacteria.Inhibitory effect was inhibited at the concentration of 15 mg/ml;The inhibitory effect of SEA on Penicillium oxalate was stronger than that of the other three.The MIC of SEA was 30 mg/ml,while that of Alternaria tenuifolia,Penicillium expansum and Cladosporium mycoides was 40 mg/ml.The inhibition effect of SEA on the growth of six bacteria was also demonstrated by the percentage of inhibition zone diameters of the two extracts to 21.3%,16.9%,15.4%,9.2%,9.5% and 10.5%.The bacteriostasis rate of SEA to four kinds of fungi was about 60%,and that of CEA to four kinds of fungi was about 50%. |
