Recently, percutaneous coronary intervention has become effective therapy to acute myocardial infarction (AMI). But, the cardiac muscle perfused by offenders vascular can't be reperfused effectively because of microcirculation disturbance. It has been indicated by variety of studies that microcirculation disturbance of coronary artery is an independent risk of AMI, so, studies on microcirculation disturbance of coronary artery while cardiac muscle ischemia-reperfusion (MIR) occured are popular, and also, it is urgent in preventing and curing MIR. Compound Danshen Pill (CDSP) is widely used in clinical because of its function of activating blood circulation to dissipate blood stasis, but, studies on the mechanism of CDSP improve microcirculation of coronary artery after MIR are rare. In our study CDSP was used to cure MIR rats, and, the microcirculation change of coronary artery, oxidative damage and endothelial cell function was observed.Methods: 60 rats were randomly divided into 3 groups:sham operated group (n=20, normal saline 1ml/d), MIR model group (n=20, normal saline 1ml/d) and MIR+CDSP group (n=20, CDSP 81mg·kg-1·d-1). The MIR model was built, and then the atorcastatin group were treated with atorvastatin for 4 weeks.then the changes of hemodynamic parameters was measured, The numbers of myocardial capillaries perfused with ink was observed, and content of plasma endothelin (ET), contents of serum maloncliddehyde (MDA) and nitric oxide (NO), and acyivity of superoxide dismutase (SOD) were detected, and correlation analysis was done.Results: Comparing with sham operated group, the numbers of myocardial capillary perfused with ink at the original ischemic place in MIR model group decreased obviously (P<0.01). The activity of SOD and the content of NO decreased obviously, but the content of MDA and ET increased significantly (P<0.05 or P<0.01). Comparing with MIR model group, the numbers of myocardial capillary perfused with ink at the original ischemic place in CDSP group increased obviously (P<0.01), the hemodynamic parameters were improved in CDSP group (P<0.05). The activity of SOD and the content of NO increased obviously whereas the contents of MDA and ET decreased obviously (P<0.05 or P<0.01).Conclusion: CDSP can improve the disorders of coronary microcirculation in MIR rats effectively, the hemodynamic parameters were improved and have protective effect on myocardium with MIR by lessening lipid peroxidation in myocardial cell membrane and enhanceing antioxygen capability of myocardial cell and adjusting equilibrium function of vascular endothelial cell etc.
