Objective: As advanced radiation therapy technology, proton radiatiotherapy has the merit to compare with other radiotherapy method in the treatment of tumors. But radiation therapy will bring some of the side effects, such as damage to normal tissue, reduced immunity etc. This paper mainly by detecting T cell and NK cell subgroups changes of cancer patients before and after proton radiatiotherapy treatment, to study the proton radiotherapy affects on cellular immune function. Methods: Choose twenty tumour patients confirmed by histopathology, 7 cases of head tumor, 8 cases of liver cancer, Other Site tumors (lung cancer, pancreatic cancer, etc.) 5 cases, 16 cases of male and female 4 cases, 24 to 73 years old, with an average age of 47.were subjected to proton rays outer local reginal therapy. Were taken before and after treatment in the peripheral blood of patients, Citrate satisfied that the anti-coagulation, Using flow cytometry detection level of T-cell subgroups and NK cells, American BD companies provide matching reagent. Results: Use statistical package SPSS14.0 statistical data processing, two groups compare use pair the t examination, the multi- groups compare use the single factor variance analysis, P<0.05was considered has statistics significance .the results showed that:CD4~+T cells,CD8~+T cells and NK cells had no significant changes, CD4~+T/CD8~+T decreased significantly after treatment than before treatment (P< 0.05). Various targets have not obvious difference between disparity in age and sex,the liver cancer patient and other spot tumor patient's CD4~+T cell has the obvious difference around the treatment. Conclusions: Tumorimmune response is mainly cellular immune response, T cell subsets is the main role, CD4~+T/CD8~+T ratio can be a direct reflection of the cellular immune function, proton radiotherapy in the treatment of cancer patients can lead to CD4~+T/CD8~+T lower, that proton therapy can bring side effects, the cellular immune function subject to certain inhibition. Is bigger to the liver cancer patient's CD4+T cell influence,but influence not obvious difference which to the disparity in age and sex.
