The expression of Homeobox(HOX) gene in the early pregnant chorion and its regulation by MifepristoneMedical abortion is considered to be a way of more convenience,less mental burden and fewer sufferings to terminate gestation.The medication of Mifepristone plus Misoprostol is most commonly used for clinical medical abortion.But it still has some disadvantages,such as long time vaginal bleeding,incompletely even failed abortion,heavy haemorrhage,secondary infection,and so on.The study on the mechanism of Mifepristone will help us better understand the process of medical abortion and the reasons for shortcomings,which will greatly improve clinical practice.HOX genes which are tightly associated to the capacity of erodibility and differentiation in tumorous tissues have obvious effects on the course of cell proliferation and differentiation.There is something in common between chorion and tumour in active hyperplasia and strong corrosion.Until now there has not any literature reported about the expression of HOX genes in early pregnant villus.This study is to determine the expressions of HOXA7,9,10 genes in early pregnant chorion and the effect of mifepristone on them.All of these may help us to know the relationship between the HOXA7,9,10 genes and the adhered and eroded capability of trophoblastic cells,which may guide us to make better choice on the dosage of medicine and weaken the disadvantages of medical abortion.The experiments here had two parts:①the expressions of HOXA7,9,10 genes in early pregnant villus;②the influence of Mifepristone on the expressions of HOXA7,9,10 genes.Part One The expression of HOXA7,9,10 genes in early pregnant chorionObjective:to determine the existence of HOXA7,9,10 genes in early pregnant villusMethods:RT-PCR was used to examine the presence of HOXA 7,9,10 mRNA in early pregnant chorionic tissues taken from 20 cases after artificial abortion and the immunohistochemistry(IHC) was performed to analyze the subcellular distribution and quantity of HOXA7,9,10 protein in 6 cases. Results:HOXA7,9,10 genes expressed in early pregnant villus,which mostly exist in trophoblastic cells and thimblefully expressed in interstitial cells.All of them are mainly expressed in cytoplasm.Conclusions:The early pregnant chorion could express HOXA7,9,10 genes.Part Two The effect of Mifepristone on the expressions of HOXA7,9,10 genesObjeetive:to understand the regulation of Mifepristone on the expressions of HOXA7,9,10genes.Methods:RT-PCR was used to detect the expressions of HOXA7,9,10 mRNA in 40 cases after Mifepristone-induced abortion(100mg Mifepristone for 20 cases,150mg for 20 cases);and the IHC was performred to analyze the subcellular distribution and quantity of HOXA7,9,10 protein in 12 samples taken after Mifepristone-induced abortion.Results:Mifepristone could significantly decrease the expressions of HOXA7 and HOXA10 gene either in the dose of 100mg or 150mg(P<0.01) and decreased the expression of HOXA9 gene without statistic significance(P>0.05).Conclusions:Mifepristone could decrease the expressions of HOXA7,HOXA9 and HOXA10 gene and which might play a role in the process of adherence and implantation in embryo.
