Experimental Research Of Anti-mammary Cancer With Homoharringtonine And Its Studies On Determination

Homoharringtonine is a kind of Chinese anti-tumor medicines.It can restrain the protein synthesis through making the ribosome depolymerized,and the number of cancer cell in patient's body drops from a high level in clinical studies and the content of nucleic acid is apparently down.Many researches show the medicine is effective for restraining the growth of different tumors,but no report about mammary cancer,so this dissertation used the method of culturing mammary cancer to study the effect of Homoharringtonine in MDA-MB-453 to find new anti-mammary cancer drugs.But to get one gram pure Homoharringtonine nearly one ton plant is need,so one kilogramme of the medicine material costs 700,000 dollars.So we also made some experiments of Determination of Homoharringtonine Injection by Capillary Electrophoresis to provide new method for appraising the content of Homoharringtonine.Results of Experimental research of anti-mammary cancer with Homoharringtonine(1)Homoharringtonine could inhibit the proliferation of breast cancer MDA-MB-543 cells in a time-dependent and dose-dependent manner.The IC₅₀ of baicalin was 92μg/ml for MDA-MB-453.(2) Trypanblau analysis showed that Homoharringtonine could inhibit the growth of cells at the doses of 10～160μg/ml for 24h,and the rate of alive cells is 98%.It shows Homoharringtonine can restrain the growth of human breast cancer cells.(3) MTT analysis showed that Homoharringtonine could increase the apoptotic rates at the doses of 10～160μg/ml for 24-72h.It shows Homoharringtonine can induce apoptosis in human breast cancer cells.(4) The morphological changes of apoptosis cells were observed under electron microscope after MDA-MB-453 cells were treated with Homoharringtonine at the concentration of 120μg/ml for 24h.Results of Determination of Homoharringtonine Injection by Capillary Electrophoresis(1) A new method for the determination of homoharringtonine in homoharringtonine injection is established by high performance capillary electrophoresis in this paper. NaH₂PO₄-Na₂B₄O₇·10H₂O buffering solution,pH=6.98,25℃,20kV working voltage,and alcohol as organic additive.With these conditions the experiment is successful. (2) Under the optimal conditions,a linearity is established between the mass concentration(X) and of peak area(Y) in the range of 0.005-0.080mg/L.Important experimental parameters including organic additive(alcohol)concentration,the types and concentrations of the buffering solution,acidity,temperature and working voltage are systematically investigated.And the results show that method is reliable and accurate.
