Objective To determine the relationships between sHLA-G expression of culture media and embryo development from Day 3 embryo transfer.Methods The expression of sHLA-G were detected by antibody-sandwich ELISA.Result(s) The positive sHLA-G expression rates of grade 1, grade 2 and grade 3 cleavage embryos from dipronucleus were 41.9%(36/86), 44.1%(49/111), 7.5%(3/40), respectively. There are no differences between grade 1 and grade 2 embryo(P>0.05), but there are significant differences in grade 3 and grade 1 as well as grade 2 embryo(P<0.05). The positive sHLA-G expression rates of 4~6 blastomeres, 7 blastomeres, 8 blastomeres and 9~12 blastomeres embryos from dipronucleus are 35.2%(13/37), 41%(16/39), 44%(40/107), 36.4%(8/22), respectively. there were no difference in subgroups(P>0.05). The sHLA-G expression rates of grade 1 cleavage embryos from dipronucleus and tripronuclear were 43.2% and 20.4%, there were significant difference between two groups(P<0.0001). The sHLA-G expression rate of blastocysts was 83.8%. The pregnancy and implantation rate in sHLA-G positive group were 68.6% (48/70) and 39.6%(57/144), whereas 10.1%(7/69) and 6.2%(9/145) in sHLA-G negative group, there were significant difference between two groups(P<0.0001). sHLA-G positive group were divided into three subgroups based on the ages, PR and IR for women≤30 years, 30~35 years and 35~38 years were 74.3%(26/35) and 42.9%(30/70), 60.7%(17/28) and 33.9%(20/59), 71.4%(5/7) and 46.7%(7/15), respectively. there were no difference in subgroups(P>0.05). Conclusion(s) The expression of sHLA-G in culture media from Day 3 has positive correlation with embryo development as well as pregnancy rate.
