| Objective To study the effects of collagen-â… sponge and chitin membrane on the proliferation ability of epidermal stem cell in vitro.And select an ideal biological scaffold material for construction of tissue-engineering skin containing epidermal stem cells.Methods Epidermal stem cells were purified by rapidly adhesion on typeâ…£collagen and amplified.The 1st generation cells were seeded into collagen-â… sponge and chitin membrane.On the collagen-â… sponge and chitin membrane,CK19 and CD29 were detected by immunofluorescence assay and the mRNA of CK19 was measured by real-time fluorescence quantitative RT-PCR after cultured for one and two weeks respectively.Results On both biological materials,the cells cultured for one week and two weeks had the expression of CK19 and CD29,and the mRNA expression of CK19 in different time had no significant difference.The mRNA expression of CK19 of epidermal stem cells on the two kinds of biological material had no significant difference.Conclusion epidermal stem Cells on the collagen-â… sponge and chitin membrane both have the expression of CK19 and CD29 in a long period of time,and the proliferation ability of cells are well maintained.Both of the materials might be used as scaffold materials for the construction of tissue-engineering skin containing epidermal stem cells. |
