Objective:To explore the high-temperature normal saline locally perfusing rabbit cerebral cortex inhibited penicillin-inducing seizure is feasible and effective.Methods:ExperimentⅠ:24 rabbits were randomly divided in 42℃,50℃,65℃,80℃group,5 rabbits in each group.The time of inhibiting penicillin-induced seizure and the impact of motor function of rabbits were observed in each group.They were respectively executed in 3 days to observe cell cortex pathology and ultrastructure changes.ExperimentⅡ:72 rabbits were divided into 3 groups(n=24):sham-operated(S) group:high-temperature(65℃) perfusion(HP) group,no reperfusion(NHP) groups, we observed the changes of each group in behavior and EEG at 0h,2h,6h,12h and measure the GABA,Glu content of the hippocampus by high-performance liquid chromatographyResults:1.The average time of 65℃group was 2 minutes to inhibit seizure.80℃group took about 1 minute,50℃group took about 12 minutes.42℃group took about 30 minutes.2.Hemiplegia took place in 80℃group,but 42℃,50℃,65℃in each group had no hemiplegia.3.Pathological changes:3.1 Change under the light microscopeEosinophilic necrosis area and nerve cell necrosis were serious in 80℃group cortical surface.We can see the phenomenon of demyelination,general subarachnoid hemorrhage,edema in deep area and some cells degeneration necrosis.Cortical degeneration necrosis and subarachnoid hemorrhage were not obvious in 65℃group, the edema can be seen only in deep zone,and no cell degeneration and necrosis;Local cortical degeneration and necrosis was not obvious,a little bit subarachnoid hemorrhage,the edema in deep zone.No cell degeneration and necrosis in 50℃and 42℃group3.2.Ultrastructural pathological changes in electron microscopy:80℃group:local neuron necrosis were seen as the most obviously in all the groups.We can see the chromatinolysis,more lysosomal,neurite spoile myelin,the vesse distention of hematoencephalic severeal;65℃group:A little local neuron necrosis appeared only in superficial layer.Hydropic degeneration happened in Chondrosomes,no neurite spoile myelin and the vesse of hematoencephalic distention was not severeal;50℃and 42℃group:We can saw the local neuron necrosis,hydrosarca in deep layer,and normal cell figure.4.Behavior and EEG observation:65℃saline local cortical perfusion significantly decreased the onset of seizures and the frequency and magnitude of epileptic discharge.The comparing during the HP group and S group,NHP group are statistical significance.(p<0.05)5.The contents of GABA and Glu in S Groups were no change in the every observation time points(OTPs).HP gourp:GABA were gradually increased in every OTPs and most obviously increase in 2h OTP.Glu content was gradully decreased and most significantly declined in 2h OTP.NHP Group:2h content of GABA and Glu did not change,6h and 12h contents of it were increased.Glu decreased slightly. The comparing during the HP group and S group,NHP group were statistical significance.(p<0.05)Conclusion:1.42℃~80℃groups local perfusion can inhibited seizrues.The higher of the temperature,the shorter of the time required for inhibiting seizure.65℃group took on average 2 minutes. 2.The higher the temperature,the more damage to the cortex,The damage in cortex caused by the 65℃saline temperature was not obvious in a short period of time.3.65℃temperature saline perfusion can inhibite the seizures and lower the frequency and magnitude of epileptic discharge.4.65℃temperature saline perfusion increased the quantity of the GABA and reduced the content of Glu in the hippocampus of the rabbitsSummary:65℃saline perfusion in rabbit cerebral cortex can safely and effectively inhibite penicillin-induced seizures.
