| Objective:Exam and compare telomerase activities in hypertrophic scar and normal skin fibroblasts.Explore the relationship between telomerase activity and hypertrophic scar pathogenesis.Methods:Collecting 11 cases of 3-6 month with 6 month or above of hypertrophic scar samples,and 22 cases of normal skin tissue.Taking 1cm×1cm×0.5cm block size of each tissue to prepare paraffin sections by immunohistochemistry;Using immunohistochemical staining method (PV-6000 two-step);Using PBS to replace the first antibody to do negative control,and breast cancer as positive control;Examine hypertrophic scar tissue by observing stains of nuclear,with HE staining-light microscopy pathological examination to determine the telomerase activity's negativity or positivity.Results:In 11 cases of 3-6 month hypertrophic scars samples,6 cases were detected positive expression of hTERT,and the positive expression rate of 54.55%;in the other hand,0 case was detected positive expression of hTERT in normal skin specimens.Statistical test:hypertrophic scar telomerase activity group and normal skin telomerase activity group,the positive rate between in those two groups was significant different(P <0.05).Conclusions:The results showed that 3-6 months of hypertrophic scar specimens hTERT positive expression rate of 54.55 percent,compared to normal skin and the difference was significant,suggesting that detection of telomerase activity can be used as a marker to predict the scar fibroblast proliferation as well as its prognosis and treatment results. |
