Systematically study on Scrophularia ningphoensis Hemsl artificial cultivated in Enshi.We study on the chemical constituent in water-soluble sector of Scrophularia ning-poensis Hemsl and get a compound.It is a new constituent isolated from Scrophularia ning-poensis Hemsl.for the first time,Named Scrophulninoside A.Identify the structure by spectroscopic methods,such as MS,1HNMR and 13CNMR,especially 2D-NMR(HSQC,HMBC).We discuss the optimum extraction of polysaccharides and pigment active part from Scrophularia ning-poensis Hemsl by orthogonal design way.Oriental water-extract way, Biological enzyme way and Ultrasonicwave way were adopted to review the content of each.We study the antifatigue function and delay-aging function of polysaccharides from Scrophularia ningpoensis Hemsl in mice and the anti-inflammatory and analgesic function of pigment from Scrophularia ningpoensis hemsl,preliminary.It shows that Compound 1 is a new constituent isolated from Scrophularia ning-poensis Hemsl.which is a iridoid glycoside in the water-abstract.By comparing three ways of extraction of polysaccharides from Radix Scrophulariae,we know the Ultrasonicwave way is better than the other two and Ultrasonicwave way is also the best way in extraction of pigment from Radix Scrophulariae.The best technic of extraction of polysaccharides is:ultrasonicwave frequency 50khz,ultrasonicwave temperature 50℃,ratio of stuff and solvent 1:25, ultrasonicwave time 90min.Under the condition,the content of polysaccharides from Radix Scrophulariae could reach 19.66%.While the best technic of extraction of pigment is:temperature 60℃,ultrasonicwave frequency 50khz,ratio of stuff and solvent 1:40,extraction time 50min.Under the condition,the content of pigment from Radix Scrophulariae could reach 21.44%.Polysaccharides from Scrophularia ningpoensis Hemsl could significantly prolong swimming time in mice,increase the reserves of liver glycogen,decreas the content of blood lactic acid after strenuous exercise.The mice gaveged the pigment from Scrophularia ningpoensis Hemsl significantly prolonged time of pain threshold and significantly reduced writhing times.In addition,the auricle inflammation and the increase of abdominal capillary permeability was significantly inhibited.
