ObjectiveIn the formation of atherosclerosis,the medial smooth muscle cell migration to the endometrium and endometrial proliferation movement is extremely important phenome -non.Balloon catheter-induced arterial injury and the occurrence of endometrial thicke -ning mechanism in smooth muscle cell membrane damage is the key.This study was to use Wistar rats artery balloon injury model of arterial injury tracking movement in mR -NA expression of smooth muscle cell membrane over time and Sivelestat sodium affec -ting the process and its related factors,to explore Sivelestat sodium affecting arterial injury of the medial smooth muscle cell migration and the level of mRNA between end -ometrial hypertrophy.MethodsSelection of 60 Wistar rats(male,12 weeks,250g±20g),their carotid artery injur -y caused by balloon catheter,were randomly divided into control group,continuous tr -eatment group,5 days treatment group and 5-days-start treatment group,a total of only four groups of treatment group,with the exception of 5-days-start treatment group,th -e other three groups were started on preoperative day dosage in the control group given n -ormal saline,only 5 days treatment group and 5-days-start treatment group continued t -o with Sivelestat sodium solution,only 5-days-start treatment delay 5 days after injury in the Sivelestat sodium solution.In addition to 5 days treatment group,which stop in only five days,and the remaining three groups stop the drug at 14 days.Four groups of rats in 1 day,3 days,5 days,7 days,rats,14days were killed.They were injected 30 min -utes before the execution by intravenous injection of 50mg/kg body weight 5-bromode -oxyuridine(BrdU).And then the application of rat pancreatic elastaseⅡcloning,in si -tu hybridization and immunohistochemical double-staining determinated arterial injury during the administration of the membrane marker BrdU elastase mRNA in arterial smooth muscle cell expression of the rate of positive cells.At the same time Intima-media thickness/diameter ratio was measured.ResultsControl group,mRNA expression of BrdU-positive cells marked rate increased sig -nificantly in 1-5 days,and then gradually declined.Continuous treatment group,5 day -s treatment group(P<0.05),only mRNA expression of BrdU-positive cells marking rate slowly increased in 1-5 days,compared with the control group statistically signific -ant differences;and sustained treatment group and 5 days treatment group,which had n -o significant statistical difference between the 5 days treatment group and 5-days-start treatment group;5 days treatment group increased in the peak of 5 days and had no sig -nificant statistical difference with the control group,.In the endometrium-media thick -ness/diameter ratio determination,the continuous treatment group and 5 days treatme -nt group only measured the ratio is lower than the control group and have significant st -atistical difference with the control group.Compared statistically with 5-days-start treat -ment group and control group,Determination of the ratio of the group had no significa -nt statistical difference.ConclusionControl group,mRNA expression of BrdU-positive cells marked rate increased sig -nificantly in 1-5 days after injury medial arterial smooth muscle cell proliferation and migration of the period can be called proliferating.Continuous treatment group,5 days treatment group,only mRNA expression of BrdU-positive cells marking rate slowly in -creased in the 1-5 days when we used Sivelestat sodium solution by inhibiting elastase, thereby inhibiting migration of medial smooth muscle cell proliferation,so that continu -ous treatment group and 5 days treatment group decline mRNA expression;and conti -nuous treatment group and 5 days treatment group which had no significant statistical difference illustrated the entire process of using Sivelestat sodium solution and drugs u -sed within 5 days after injury have the same effect,further major role was in the drug 5 days after injury,that was proliferative drug.5-days-start treatment group increased in the peak of 5 days and had no significant statistical difference with the control group,a -nd there was not obvious after the administration of proliferation.In the intima-media t -hickness/diameter ratio,the ratio of the continuous treatment group and 5 days treatm ent group measured was lower than of the control group and had statistically significant differences with the control group,so we think sivelestat Sodium can delay endometrial hyperplasia;the ratio of 5-days-start treatment group had no significant statistical differ -ence with the control group ratio,so that we think the role of endometrial hyperplasia after 5 days was not obvious when we used sivelestat sodium and sivelestat sodium gi -ven after the administration of the proliferation can not be the role of the elimination of endometrial hyperplasia.
