Objective60%-90%of primary ovarian malignant tumors comed from epithelial,and the relapse rate and mortality rate is at the first of female reproductive system tumor.At present,despite chemotherapy and radiotherapy and surgical technology continuing to improve,but ovarian cancer 5-year survival rate has not been changed significantly. Mainly because the early symptoms of ovarian cancer was not obvious,more than 70% of patients with ovarian cancer is already advanced at the time of diagnosis.Prognosis of ovarian cancer is closely related to clinical stage.The five-year survival rate of patients with early ovarian cancer is up to 80%.Therefore,over the years by experts on the early diagnosis of ovarian cancer,the pathological form of the law of continuous development of clinical research.Therefore,over the years many experts study on the early diagnosis of ovarian cancer,pathological,and the law of clinical happened ongoing.Numerous studies show that there are relationship between the mitochondrial dysfunction and tumor formation.Mitochondria is a eukaryotic cell contains the only genetic material outside of nuclear mtDNA,within the main site of energy synthesis, and involved in many pathophysiology of metabolic processes,for example tumorigenesis and aging.It also play an important role in the process of apoptosis and cell biological behavior of other signal transduction.Our preliminary work,using 2D electrophoresis compared the expression of the protein in 18 cases of ovarian serous cystadenocarcinoma and 7 cases of normal ovarian.We found the mitochondrial short-chain coenzyme A hydratase expression increased,reached 2.74-fold difference in expression.Based on the results of the preliminary work,the experiment through the reverse transcription-polymerase chain reaction to study the expression of mitochondrial short-chain coenzyme A hydratase mRNA in ovarian serous cystadenocarcinoma. Preliminary study the possible relationship between it with the occurrence and development of the ovarian serous cystadenocarcinoma.Materials and Methods75 cases including 15 cases of nomal ovarian tissue,15 cases of benign epithelial ovarian tumor,45 cases of Ovarian serous cystadenocarcinoma were obtained from patients who undergone operation for cervical cancer,ovarian tumor,and epithelial ovarian cancer from Jan 2006 to May 2008 in Shengjing hospital.15 cases of normal ovarian tissue were chosen as the normal group,15 cases of benign epithelial ovarian tumor were chosen as benign group.The tissue specimens were preserved in liquid nitrogen at -80℃immediately after surgical removal.45 cases of Ovarian serous cystadenocarcinoma included 15 cases ofⅠstage,15 cases ofⅡstage,15 cases ofⅢstage.45 cases of serous cystadenocarcinom were composed of 24 cases of low differentiation,13 cases of moderate differentiation,8 cases of well differentiation.All patients are without preoperative radiotherapy,chemotherapy and hormone therapy and informed consent.Reverse transcription polymerase chain reaction(RT-PCR) were used to detect the expression of short chain enoyl—coa hydratase mRNA in 45 cases of ovarian serous cystadenocarcinoma(cancer group),15 cases of ovarian serous cystadenoma(benign group) and 15 cases of nomal ovary tissues(normal group).All data were expressed as mean±standard deviation((?)±s).SPSS version 14.0 was used to performed statistical analysis,and the differences in data between group were analysis using LSD-test,t-test and one way ANOVA.P<0.05 was considered statistically significant.ResultsThe mRNA of short chain enoyl—coa hydratase were expressed in normal ovarian tissues,benign epithelial ovarian tumors and epithelial ovarian cancer.The expression of the mRNA which encode short chain enoyl—coa hydratase were significantly higher in ovarian serous cystadenocarcinoma than that in benign epithelial ovarian tumors and normal ovarian tissues(p<0.001);Its expression was no significant difference in benign group compared with the normal group(p>0.05).The overexpression of the mRNA which encode short chain enoyl—coa hydratase were no relationship with the clinic stage and histological grade(p>0.05).ConclusionThe mRNA of short chain enoyl—coa hydratase were expressed in ovarian serous cystadenocarcinoma,ovarian serous cystadenoma and normal ovarian tissues.Its expression in the ovarian cancer group was significantly higher than that in benign group and the normal group;and was not related to clinicopathological parameters.
