ObjectiveSearch for microRNAs associated with ovarian serous carcinoma,and to accumulate the data of microRNA expression in patients with ovarian cancer.Furthermore,it can provide a theoretical and practical basis for the early diagnosis and treatment of ovarian cancer.MethodsThe specimens were randomly selected in 3 cases of serous ovarian cancer tissues and 3cases of normal ovarian tissue,using Agilent Human microRNA(8*60K)V21.0 chip(covering 2549 human related microRNA),preliminary screening of ovarian serous carcinoma showed a differential expression of microRNA.According to the results of the microRNA chip,the real-time quantitative reverse transcription PCR was used to validate the 58 ovarian tissue samples.The relationship between microRNA expression and clinicopathological features of ovarian serous carcinoma was analyzed.ResultsCompared with the normal ovarian tissue,nine microRNA were up-regulated in ovarian serous carcinoma by microarray,they are hsa-miR-141-3p,hsa-miR-200a-3p,hsami R-200b-3p,hsa-miR-200c-3p,hsa-miR-224-5p,hsa-miR-7-5p,hsa-miR-142-3p,hsa-miR-21-5p,hsa-miR-142-5p,respectively.Six microRNA was significantly decreased,they are hsa-mi R-424-5p,hsa-miR-214-3p,hsa-miR-125b-5p,let-7b-5p,hsa-miR-199a-5p,hsa-miR-100-5p,respectively.The results of real-time fluorescence quantitative PCR are consistent with the chip.And in the stratified analysis of different microRNA,compared with no lymph node metastasis and I-II stage,hsa-miR-21-5p expression increased in III-IV stage lymph node metastasis patients,and hsa-miR-125b-5p expression decreased,the differenceswere statistically significant(P < 0.05);but there was no significant difference between high level and low level in ovarian cancer patients(P >0.05).ConclusionsThe expression profile of microRNA gene in ovarian serous carcinoma was preliminarily established,and 15 differentially expressed microRNA were screened out.Real-time quantitative reverse transcription PCR technique was used to verify the differentially expressed genes.The abnormal expression of hsa-miR-21-5p and hsa-miR-125b-5p in ovarian serous carcinoma may have prognostic significance.
