Study On The Effect Of Treatment Of Invigorating Spleen And Promoting Blood Regulating Qi On PAI-1 U-PA And PⅢNP Of The Hepatic Fibrosis Rats

Objective:To establish a rat model of liver fibrosis,Research under the guidance of Shugan Jianpi Huoxue method used Chaihu Guizhi Tang on liver fibrosis Rat liver cells PAI-1,u-PA,Pâ…¢NP.It aimed Depth study of the method in the treatment of liver fibrosis in rats.Method:60 male Wistar rats,were randomized into 5 groups:Model group,low dose group of Chinese medicine,Chinese medicine high-dose group and blank control group of western medicine group.Made for the use of CCl₄ composite model,model 2 weeks after the low dose group of Chinese medicine,Chinese medicine began to use high-dose group administered Chaihu Guizhi Tang treatment,western medicine,the control group treated with oral colchicine.Rats were sacrificed after eight weeks,and was detected by immunohistochemistry in rat liver tissue PAI-1,u-PA expression;using enzyme-linked immunosorbent assay in serum Pâ…¢NP content.Results:1.Liver tissue of PAI-1 protein expression Low-dose group of Chinese medicine,Chinese medicine high dose group the expression of PAI-1 compared with the model group significantly lower(P＜0.01)2.Liver tissue of u-PA protein expression Low-dose group of Chinese medicine,Chinese medicine high dose group the expression of u-PA compared with the model group increased(P＜0.01)3.Serum Pâ…¢NP content Model group,serum Pâ…¢NP content of the highest,low-dose group of Chinese medicine,Chinese medicine high-dose serum Pâ…¢NP in a significant reduction in the content(P＜0.01)Conclusion:Chaihu Guizhi Tang in inhibiting PAI-1 expression in the liver,at the same time it can enhance the expression of u-PA,to accelerate the degradation of collagen plays the role of anti-hepatic fibrosis;Chaihu Guizhi Tang can reduce the serum concentration of Pâ…¢NP,through the inhibit the synthesis of collagen typeâ…¢,thereby reducing the deposition of ECM and the degree of liver fibrosis.
