Objective:To study the changes of serum AMH,FSH,E2 and ovarian histological in rats of cisplatin-induced premature ovarian failure in order to investigate serum AMH,FSH,E2 which can be assessed to the early prediction indicators of ovarian reserve in adult female rats,so as to study the human premature ovarian failure and provide a theoretical model for early diagnosis of premature ovarian failure and reliable predictors.Methods:Adult female SD rats were treated with saline,4.5mg/kg and 6.0mg/kg cisplatin by intraperitoneal injection to establish successfully in rat model of chemotherapy induced premature ovarian failure.The serum AMH,FSH,E2 were measured by using an Enzyme-linked Immunosorbent Assay.The number of original,primary,secondary,mature and atresic follicles were counted by using paraffin sections under microscope in the largest ovarian cross-section.Vaginal smear record changes in the estrous cycle.By using SPSS11.5 statistics package to finish statistical analysis of data.Results:Estrous cycle of different doses of cisplatin group was obviously longer than the control group in rats after intraperitoneal injection.And it took on a dose-related change.The serum AMH levels were significantly lower in both cisplatin groups.The serum E2 levels were significantly higher in low-dose group but there were not significantly in high-dose group compared with the control group.The serum E2 levels were significantly lower in high-dose group compared with the low-dose group.The serum FSH levels were not significantly in low-dose group compared with the control group but significantly higher in high-dose group compared with low-dose group and control group.Estrous cycle of high-dose group after intraperitoneal injection was obviously longer than the non-injection.The serum AMH levels were significantly lower in both cisplatin groups after injection than before the injection.The serum E2 levels were significantly higher in low-dose group but there were not significantly in high-dose group after injection than before the injection.The serum FSH levels were significantly higher in both cisplatin groups after injection than before the injection.The number of original and primary follicles was significantly reduced in the low and high dose group and the number of atretic follicles was significantly increased in a dose-related change after injection.Conclusions:1.Cisplatin can lead to chemotherapy induced premature ovarian failure in rats.2.Cisplatin-induced ovarian damage in the early rise of serum E2 compensatory and AMH decreased obviously.These two hormones changes as early as FSH.3.The rise of serum E2 and decreasing serum AMH can be used as the biomarkers of ovarian damage after exposuring to chemotherapy for early predictor,and two together will help the early diagnosis of premature ovarian failure.
