Liposome encapsulating with vaccine has the function of adjuvant and cartier. Drug-loaded liposome is an ideal immune adjuvant,after they entered the body, majority of them were uptook by phagocytic cells,and easily focused on the main immunological organ,which is good for delivering the antigen to antigen-presenting cells or other immunological activated cells to activate and enhance the immune response.Pulmonary delivery of liposome vaccine not only has the obvious function of adjuvants to enhance immune response,but also the inductive effect of mucosal,cellular and humoral immunity.Using phospholipids and cholesterol as raw materials,this research employed the thin flim evaporation-lyophilization method to produce dry power of purified influenza virus split vaccine liposome.The main influence factors were investigated in the preparation process of liposome suspension and the freeze-dried powder. Taking encapsulation rate which determined by Lowry method as index,we selected the best prescription for liposome suspension by the uniform design method,the best freeze-drying technique and the protectant dosage through the single-factor study.We also analyzed the particle shape and size distribution through microscopic observation, studied the liposome suspension and dry powder's prilimanary stability by taking appearance and encapsulation rate as index.In this study,the traditional injection approach of vaccination immunity was changed to pulmonary vaccine delivery to Clean ICR mice,followed by the liposome vaccine immunogenicity investigation through the antibody titles which determined by micro-determination of HI method.According to the optimized prescription,the encapsulation rate of influenza vaccine liposome lyophilization powder was 84.78%,the liposome particles were in forms of round or elliptic,at a mean size of 2.14μm,distributed evenly,performed well in immunogenicity and stability;Compared with intraperitoneal injected mice,at the same dose,the mice which took pulmonary delivery of influenza vaccine produced earlier antibodies,higher antibody level and stronger immune response.
