| Purpose:To investigate the in vitro effects of celecoxib, HA and compound of celecoxib and HA on rabbit chondrocytes cultivated for 24h in the presence of interleukin-1β(IL-1β),and clarify the relevant mechanisms of OA treatment.Methods:New Zealand rabbit chondrocytes were cultured in vitro .Then all the chondrocytes were divided into five groups: negative group, positive group, HA group,celecoxib group and compound of celecoxib and HA group.Negative group was the normal chondrocyte,postive group was the chondrocyte stimulated by IL-1β,the other three experiment groups were added the three drugs respectively after stimulated by IL-1β.After 24 hours the chondrocytes were collected to detect the MMP-1,MMP-3 and TIMP-1 gene expression.All the medium were collected to detecte tumor necrosis factorα(TNF-α) and prostaglandin E2 (PGE-2).Results:In the experiment group the MMP-3 gene expression in the chondrocytes was lower than the postive group,higher than the negative group,the differences were both significant(P <0.05).Meanwhile in the three experiment group, TNF-αand PGE-2 which chondrocytes released to the medium were lower compared with postive group(P <0.05). Among the three groups there were no significant differences(P>0.05).Conclusion:All the three drugs prevented release of PGE-2 and TNF-αfrom chondrocyte,then alleviated inflammation.The three drugs also inhibited the expression of MMP-3,then preventing the chondrocyte matrix degradation. The mechanism of the three drugs treatment of early osteoarthritis was to inhibited the MMP-3, PGE-2 and TNF-αexpression. |
