Objective:This study was performed to elucidate whether these antigen-processing gene(LMP/TAP)polymorphisms could influence the infection of Tuberculusis in the Li population in hainan in China.Methods:Casecontrolstudydesi gnwasado pted.Atotal of 205 welldefine pulmonary Tuberculusis patients and 217 unrelatednormal controls werereruited for thestud. Genomic DNA of 205 Tuberculusis(TB)patients and 217 healthy controls were extracted from the Li population in hainan in China.Polymorphisms of LMP/TAP genes in Tuberculusis were determined by polymerase chain reaction-restriction fragment length polymorphism(RFLP),the controls by DNA sequencing.We use the software PHASE1.0 to construct the haplotypes of every individual.At last the unconditional Logistic regression model was used to analyze the statistical association of genotypes or haplotypes in two groups adjusted by gender and age.Results:The distributions of LMP2 genes between cases and controls did not differ.However,LMP7 gene frequency in patients was higher than that in controls odds ratio 2.97(95%confidence intervall.80-4.90);3.77(95%confidence interval 1.60-8.89),heterogenous or homologous respectively].Similarly;The distributions of TAP1 Codon333 genes Ile/Ile→Ile/Val frequency in patients was higher than that in controls odds ratio P=0.008,OR=1.92(95%CI:1.19-3.12),but Ile/Ile→Val/VaL between cases and controls did not differ(P>0.05).There are significant differences in the SNPs of Codon637 between the healthy control and Tuberculosis patients in Li population Hainan in Chinese:Asp/Asp→Asp/Gly P<0.01,OR=2.87 (95%CI=1.75-4.71),Asp/Asp→Gly/Gly OR=3.94,(95%CI=1.82-8.53).Similarly,we find the combination of-B/+B/B,--/H +H/H/,--/G+G/G,--/I+I/I has a significant difference in two groups:OR=3.674 P<0.0001 95%CI[2.254-5.9888];OR=3.573 P=0.009 95%CI[1.379-9.263];OR=0.334 P=0.043 95% CI[0.115-0.968];P=0.034 OR=2.909 95%CI[1.084- 7.807]。Conclusion:These findings suggest that polymorphism of LMP7 gene is one of the important host factors which independently influence the infection of Tuberculusis,and the risk of Tuberculusis in participants with Gln/Lys allele is 2.97 and Lys/Lys allele is 3.77-fold compared to those with Gln/Glnallele,in the Li population in hainan in China.;SNPs at Codon333 of the TAP1-1 gene may relate to the susceptibility of Tuberculusis,and the risk of Tuberculusis in participants with Ile/Val allele is OR=1.92;SNPs at Codon637 of the TAP1-2 gene may relate to the susceptibility of Tuberculusis,and the risk of Tuberculusis in participants with Asp/Gly allele is OR=2.87 and Gly/Gly allele is 3.94-fold compared to those with A/A allele.These findings suggest that SNwhich independently influence the occurrence of Tuberculosis.Furthermore compared with other haplotypes,--B/+B/B,--/H +H/H can rise the risk of Tuberculusis 3.674 and3.537-fold.These findings suggest that SNPs of LMP/TAP gene is one of the important host factors which independently influence the occurrence of Tuberculosis.
