1 Experiment of establish rats model with diabetic nephropathyObjective To establish models of diabetic nephropathic rats,and to observe their general condition, Levels of podocalyxin expression ,levels of blood glucose,Quantities of 24 hours urinary protein excretion, serum creatinine,urea nitrogen and pathological changes of renal tissue at corresponding time points.Methods 45 male wistar rats were randomly divided into three groups of control group, diabetic nephropathic model group for 4 weeks and 12 weeks,each group have 15 rats. Then rats of model groups have been treated by streptozotocin (STZ) after there right kidney have been excised.Levels of blood glucose,quantities of 24 hours urinary protein excretion were examined at the end of 2,4,6,8,12 weeks. Rats in model group for 4 weeks were sacrificed at the end of 4 weeks,and rats in control group and model group for 12 weeks were sacrificed at the end of 12 weeks respectively.Then levels of podocalyxin expression,Serum creatinine (Scr), blood urea nitrogen (BUN),pathological changes on renal tissue have been examined and observed.Results Levels of blood glucose and quantities of 24 hours urinary protein excretion were higher in model groups than that of control group at the end of 2, 4, 6, 8 and12 weeks. At the end of 4, 12 weeks, podocalyxin expression in model groups were lower than control group(P<0.05). Levels of podocalyxin expression in renal tissues of model rats for 12 weeks' group were lower than in group of 4 weeks(P<0.05).levels of BUN and Scr in this group were lower than in group of 4 weeks(P<0.05).Further more, pathological changes such as thickening of glomeruler basement membrane and proliferation of mesangium matrix that characterise the diabetic nephropathy has been found in model group at the end of 4 weeks.And,these changes were much more severe in renal tissues of rats at the end of 12 weeks.Conclusion Rats model of diabetic nephropathy had been successfully prepared and the foundation for study of drug intervention on these model had been settled. 2 Experimental Study on Effect of Shenyankangfu Tablet on Podocalyxin in Renal Tissue of Rats with Diabetic NephropathyObjective To investigate the effects of Shenyankangfu Tablet(SYKF) on expression leves of podocalyxin in renal tissue and blood glucose, quantities of 24 hours urinary protein excretion , creatinine,urea nitrogen and pathological changes of renal tissue of rats with diabetic nephropathy.Materials and Methods Sixty male wistar rats were randomly divided into the following four groups: control group (n=15),diabetic nephropathic model group (n=15),SYKF–treated group (n=15), and losartan–treated group (n=15). The DN- models were induced by method recommended above. Two days after models have been prepared, rats were fed with shenyankangfu tablet (600 mg·kg - 1·d - 1) in SYKF–treated group,and were fed with losartan(20 mg·kg - 1·d - 1) in losartan–treated group.rats in both control and model group were were fed with same volume of water. Levels of blood glucose,quantities of 24 hours urinary protein excretion were examined at the end of 2,4 ,6,8,12 weeks. All rats were sacrificed at the end of 12 weeks.Then levels of podocalyxin expression , Serum creatinine (Scr), blood urea nitrogen (BUN),pathological changes on renal tissue have been examined and observed.Results Levels of blood glucose and quantities of 24 hours urinary protein excretion were in normal range at the end of 2,4,6,8,12 weeks in control group. expression levels of podocalyxin , Scr and BUN of this group were in normal range, too, at the end of 12 weeks.All these indexes were significantly higher in model group and two durg treated groups than in control group, respectively (P < 0. 05),but Expression levels of podocalyxin was lower than control group(P < 0. 05) . At the end of 12 weeks, expression levels of podocalyxin in drug treated groups were higher than model group(P < 0. 05). Pathology changes is less severed than model group. Levels of blood glucose in these groups were a little low than model group,but have no statistical significance(P>0.05)。At the same time, quantities of 24 hours urinary protein excretion, Scr and BUN levels were lower in drug treated groups than in model group. (P<0.05).Conclusion Shenyankangfu tablet can up regulate the expression of Podocalyxin on renal tissue of rats with diabetic nephropathy ,decline levels of quantities of 24 hours urinary protein excretion of different stage ,and levels of Scr and BUN at the end of 12 weeks. to ease pathologycal changes,and have protect effects on on renal tissue of rats with diabetic nephropathy.
