Objective:To detect the expression of WT-1 protein,Anti-Mullerian Hormone and Ki-67 in ovarian granulosa cell tumour.And to explore the expression of WT-1 protein and Anti- Mullerian Hormone in the development of ovarian granulosa cell tumour and the relationship between them.To explore the role of WT-1 protein and Anti-Mullerian Hormone in the development of ovarian granulosa cell tumour,and to study the relationship between the expression of them and the bionomics of ovarian granulosa cell tumour.Besides,we try to explore the value of tumour's differentiation in predicting the prognosis of ovarian GCT.Methods:37 cases of the ovarian granulosa cell tumour and 10 cases of normal ovary were detected on Tissue Microarries,the expression of WT-1 protein,AMH and Ki-67 was measured with Immunohistchemistry.The results were analyzed with SPSS 11.5 statistical software.Results:①In the normal ovary,every index only express in granulosa cells of primary follicles and secondary follicles,but has no expression in the primitive and mature follicles.The ratios of hypo-expression of WT-1 protein and AMH between the primary follicles and the secondary follicles were significantly different(P<0.05).The expression of Ki-67 in primary follicles and secondary follicles has no significant difference(P>0.05).The ratios of expression of WT-1 protein and AMH in ovarian granulosa cell tumour are 81.1﹪and 78.4﹪.②The ratios of hypo-expression of WT-1 protein and AMH in the recrudescent patients and the patients with ascites were lower than that in the unrecrudescent patients and the patients without of ascites,and the difference has statistical significance(P<0.05).The proliferation index of Ki-67 has nothing to do with recidivation and ascites,but it was concerned with lymph node metastasis(P<0.05).Besides,the ratios of hypo-expression of WT-1 protein and AMH in tumours of which the histotype is asystematic,in the cases of caryocinesis≥3/10HPF and the tumours with conspicuous heteromorphism of cells were lower than that in tumours of which the histotype isn't asystematic,in the cases of caryocinesis<3/10HPF,and the tumours without of conspicuous heteromorphism of cells,and the difference has statistical significance(P<0.05).The proliferation index of Ki-67 were also concerned with the tumour's histotype,the tumour cells' caryocinesis and heteromorphism(P<0.05).③The ratios of positive expression of WT-1 protein and AMH in stageⅠw ere higher than that in stageⅡa nd stageⅢ(P<0.01).The proliferation index of Ki-67 was also concerned with the clinical stage(P<0.01).④The tumour's histotype,the tumour cells' caryocinesis and heteromorphism were concerned with the tumour's recidivation and clinical stage.In the patients of stageⅠ,the ratio of the recrudescent patients in tumours of which the histotype is asystematic,in the cases of caryocinesis≥3/10HPF and the tumours with conspicuous heteromorphism of cells were higher than that in tumours of which the histotype isn't asystematic,in the cases of caryocinesis<3/10HPF,and the tumours without of conspicuous heteromorphism of cells.and the difference has statistical significance(P<0.05).In the patients of stageⅡand stageⅢ, the tumour's recidivation was nothing to do with the tumour's differentiation.⑤The expression of WT-1 protein was positively correlated with AMH (r=0.819,P<0.01).Conclusions:①Detection of the expression of WT-1 protein,AMH and Ki-67 can determine the biological behavior of ovarian GCT and help us to know the development of the tumour.②T he expression of WT-1 protein and AMH is concerned with the development of ovarian GCT.③Differentiation of the tumour can be used as a referred factor to evaluate the prognosis of ovarian GCT.
