| Malpighia glabra,which has the highest vitamin C content of fruit in all existing types of fruit.And it attracts more and more attention for its high nutritional value and natural vitamin C content.The current studies of Malpighia glabra are limited to the introduction and cultivation of Cherry and food and beverage processing,and a little about the Malpighia glabra processed freeze-dried powder.Malpighia glabra freeze-dried powder in cosmetics applications,both at home and abroad have not been reported.This subject is the first time comes from the point of cosmetic application,which conducts a comprehensive,integrated research and development and utilization of the Malpighia glabra extract of freeze-dried powder,and provides a theoretical basis for the development and utilization of the Malpighia glabra resources and the industrial development of its effective products.This subject studied in the Malpighia glabra extract of freeze-dried powder and its extraction procession,determined the optimum liquid ratio,extraction time, temperature,the main components;carried out the desalination treatment;did the efficacy of the antioxidant solution,whitening effect and a stimulating researches;and finally formed a stable prototype system of deoxidized vitamin C in the extract.The results as follows:1.The best extraction conditions ofMalpighia glabra extract as follows:liquid ratio of 1:10,under the conditions of stirring at room temperature for 1 hour extraction. The extract deoxidized vitamin C content in the extrac after centrifugational filtration is 10±0.3mg/ml,polysaccharide content is 25.3mg/ml,protein content is 1.02mg/ml,low relative content of protein,of which the content of nicotinamide is about 46.8mg/L.2.The ultimate selection of an appropriate resin of the desalination extract,which is cationic resin T212,anion resin T112,filter size reached of 42 columns,loss rate of Vc in the process of desalination was 14.2%,which provided data support for the industrial production.3.The use of different free radicals to carry out a study in antioxidant activity of extract liquid.The results showed that when 20-fold diluted(Vc content of the extract is 500μg,)the extract of superoxide anion radical scavenging rate was 90.3%;50 times diluted extract(Vc content of the extract is 160μg),the hydroxyl radical scavenging rate was 92.2%;when extract concentration of 40μg/ml in Vc, DPPH.clearance rate were over 90%.In addition,with the same concentration of extract compared to the Vc,it has a higher capacity in the free radical scavenging,which indicating that not only Vc at work in the extract,but also contains other active ingredients,such as nicotinamide,which is the combined result of the various components.4.Through Enzyme assay of biochemical to detect the inhibitory effect of the extract on the activity of tyrosinase.The results showed that:in the 2.5%extract concentration that is reduced Vc content 0.25mg/mL tyrosinase activity on the inhibition rate reached 99.1%.5.By the Experimental Detection of animal cell to detect the inflence of the extract on the Proliferation inhibition rate of mouse B16 melanoma cells and Melanin content of cells and determination of viability of cell tyrosinase.The results show that the,MTT results show that different concentrations of the extract had no effect on the basic dynamic of B16 melanoma cells,was able to reduce the melanin content,decreased tyrosinase activity,could inhibit cell mRNA expression of tyrosinase,compared with the pure Vc,the extract had a more obvious whitening effect.As the effect of expression of inhibition on tyrosinase mRNA by Vc was not obvious,which also showed that other components of extract inhibited the expression of tyrosinase mRNA,there by reducing the generation of melanin.6.The results of Human vitro Whitening test had confirmed the extract can reduce skin melanin content,and increase the L value.But not the higher the concentration the better,there is the issue of skin absorption maximum.Different concentrations of the Malpighia glabra extract of freeze-dried powder of the hemolytic rate is lower than 5.0 percent,which show extract is without injury of the cell,without proof of a strong extract irritant.7.By using the red blood cell hemolysis test to detect the irritation of the extract on the membrane.The results showed that the hemolytic rate of different concentrations of the Malpighia glabra extract of freeze-dried powder were all lower than of 5.0 percent,which showing no injury to the membrane and no strong irratant.8.To establish a relatively stable system of extract,that is,using the deionized aqueous which has been boiled and then cooled to room temperature,liquid ratio of 1:10,under the conditions of stirring at room temperature for 1 hour extraction. Added 30%(mass ratio)of polyol to Centrifugal filter extract,uniformly mixed, and save aider sterilization.In the above test,the optimization of the Malpighia glabra extract of freeze-dried powder process verified that the extract has the effectiveness of anti-aging cosmetics,whitening,and also built a relatively stable system of the extract,which provided a theoretical and data support to the development and utilization of resources of the Malpighia glabra. |
