ObjectiveChemerin is recently found as a chemoattractant.It can promote chemotaxis of dendritic cells and macrophages though binding to its receptor ChemR23.We have proved Chemerin-encoded gene TIG2 is a differentially expressed gene of lung cancer. Motion ability of A549 cells enhanced after recombinant plasmid of Chemerin transfected into A549 cell line.We detected the expression of Chemerin in peripheral blood of patients with lung cancer and its effect on in peripheral blood of patients with lung cancer and its effect on NF-κB and JNK signaling pathway in A549 cell line in this study.Methods1.The concentration of Chemerin in peripheral blood of 42 patients with lung cancer and of 31 health examinations were detected by the ELISA double sandwich method.T test and one-way ANOVA were used to analyze between the concentration of Chemerin in peripheral blood of patients with lung cancer and the relationship with clinicopathological factors.2.Lung adenoma cell line A549 was cultured in vitro,and was processed by recombinant Chemerin of different concentration level.After cell lysed,we used western blot method to test the expression of phospho-P65 and phospho-JNK in groups of different concentration level.One-way ANOVA was used for statistical analysis.ResultsThe concentration of Chemerin in peripheral blood of 42 patients with lung cancer (1773.21±328.37pg/ml) were significantly higher than the concentration of 31 health examinations(1373.72±411.62pg/ml),p<0.001.The concentration of Chemerin in peripheral blood of patients with lung cancer had no relationship with clinicopathological factors,p>0.05.Western blot result showed that the expression level of phospho-P65 increased in A549 cell line processed by Chemerin,and the expression level of phospho-P65 demonstrated a positive correlation with the concentration of Chemerin.Meanwhile,the expression level of phospho-JNK decreased in A549 cell line processed by Chemerin,and the expression level of phospho-JNK demonstrated a negative correlation with the concentration of Chemerin.Conclusions1.The concentration of Chemerin in peripheral blood of patients with lung cancer is higher than the concentration of normal persons,p<0.001.2.The concentration of Chemerin in peripheral blood of patients with lung cancer has no statistical relationship with clinicopathological factors.3.It shows a concentration-dependence between the NF-κB pathway activation/ JNK pathway inhibition and recombinant Chemerin process to lung adenoma cell line A549.
