Quercitrin-3-glucoside (QG) is a kind of flavones monosomic compounds, which is a derivate, its glucoside on 3 posite of quercitrin. It widespreadly exist in natural plants. Literature has reported that quercitrin, mother nuclide of QG, has protective effects on myocardial ischemia injury. But till now, there is no report about the effect of QG on myocardial ischemia injury. In this study, the protective effect of QG on myocardial ischemia injury and its mechanism were studied. Results were showed as followed:1. QG improve acute myocardial ischemia injury induced by isoproterenol in miceOn the model of myocardial ischemia mice induced by Iso, QG 5, 10mg·kg-1 markedly inhabited Iso-induced the raise of the ST segment of ECG and the increase of MDA in myocardium, raised the activities of LDH and GHX-Px; QG 2.5, 5, 10mg·kg-1 remarkably raised the activity of SOD in myocardium.2. QG prolonged the ECG time after clamping trachea in miceOn the model of myocardial anoxia induced by clamping trachea in mice, QG 5, 10 mg·kg-1 significantly prolonged the ECG time after clamping trachea.3. Protective effect of QG on the myocardial ischemia-reperfusion(I/R) injury induced by occluding the left anterior descending (LAD) coronary artery in miceAfter ligating LAD coronary artery in mice, the ST segment and T wave of ECG were markedly raised, LDH activity and MDA content in serum were significantly increased, while myocardial infarction was produced remarkably. QG 20 mg·kg-1 obviously reduced the height of ST and T segment after occluding 5min; QG 10 mg·kg-1 obviously reduced the height of ST and T segment after occluding and reperfusion 30min; QG 10, 20mg·kg-1 inhibited the increase of MDA content and LDH activity in serum; QG 10, 20mg·kg-1 significantly reduced infarction area.4. Protective effect of QG on the myocardial I/R injury induced by occluding the LAD coronary artery in ratsOn the model of myocardial I/R injury in rat, QG 3.5, 7, 14 mg·kg-1 significantly inhibited raise of ST segment after occluding 2min; QG 7, 14 mg·kg-1 obviously inhibited raise of ST segment after occluding 30min; QG 7, 14 mg·kg-1 markedly inhibited raise of ST and T segment after reperfusion 30min; 7, 14 mg·kg-1 QG significantly reduced increase of LDH activity and MDA content in serum, while remarkably raised the activity of content of NO; 3.5, 7, 14 mg·kg-1 QG significantly reduced infarction area.Conclusions: QG has a protective effect against myocardial ischemia or I/R injury in mice and rats, the protective mechanisms might be related to attenuating oxygen free radicals and increasing of NO production.
