Aim Erythropoietin(EPO) is known as a hematopoietic growth factor,which has been used to treat anemia caused by different reasons.But recent evidence demonstrated several non-haematopoietical effects including anti-apoptotic effects and pro-angiogenic effects which could be of myocardial protection in patients with ischemic heart disease. These effects may be involved the therapeutic bases for the treatment of cardiovascular diseases.This study was designed to observe the myocardial protective effect of the erythropoietin(EP0) after acute myocardial infarction of rats.Methods The model of acute myocardial infarction of rats use the way of standardized rat model of myocardial infarction by ligating the left anterior descending coronary artery after thoracotomy under the conditions of general anesthesia,non-invasive respiratory support.The model of acute myocardial infarction of rats were randomly divided into two groups,EPO group and the control group,3000u/kg EPO was intraperitoneally injected into the rats EPO group,3 days in total(1 day before operation,1 the day of the operation,1 day after operation ).The rats of control group were given the same volume of normall saline at the same each time piont.After 7 days in each group were randomly selected five rats and haemospasia from tail vein of the rats,then deploy the method of ELISA to detect the contents in blood serum of TNF-α, IL-6.After 28 days of operation,arterial cannula through the right common carotid artery of rats and detect the index of haemodynamics,then deploy immunohistochemistry ofⅧfactor and the Bcl-2,Bax of apoptosis protein.Results In the experimental process,a total of 7 deaths,one of the death in the course of anesthesia,2 rats because of chest wall vascular injury in hemorrhagic shock led to death,one of the death because of malignant arrhythmia after anterior descending artery ligation,There are three rats death because of heart failure,lung infections and other reasons after surgery.Immunohistochemical experiments showed thatm,compared with control group,the level of TNF-αand IL-6 was obviously degraded,the expression of the Bcl-2 of apoptosis protein was up-regulation,the Bax was down -regulation and the ratios of Bcl-2/Bax was obviously increased in EPO group(P<0.05).The capillary density of myocardial infarction peripheral zone of rats was obviously increased (6.3±0.7,4.6±0.9,P<0.05).The hemodynamic measurement of LVSP,+dp/dtmax were improved and the LVEDP,-dp/dtmax were decreased in EPO group(P<0.05).Conclusion Approve the granting of erythropoietin in rats after acute myocardial infarction,its can improve the cardiac function of acute myocardial infarction of rats through the effects promote the formation of capillary in the surrounding region of infarction,inhibition inflammatory response in the surrounding area infarction and reduced cardiomyocyte apoptosis.
