Pulmonary fibrosis (PF) is a kind of serious lung diseases characterized by diffuse progressive fibroplasias and alveolar structure which finally leading to disorder of lung function, and is the common final of many lung diseases. The exact pathogenesis of pulmonary fibrosis is not thoroughly understood.The destruction of alveolar epithelium barrier, inflammatory cell infiltration and disequilibrium on the cytokine-network, and subsequent collagen accumulation may be the common pathway in different types of pulmonary fibrosis.The etiological factors of PF are diversity, and there is no satisfactory therapy for it up to now. Glucocorticoid and cytotoxic drug are the first choices in treating PF in clinic, but the therapeutic efect is indefinite accompanied with obvious side effect. So the urgent question which the scholars need to solve is to seek effective and safe medicine for treating PF.Traditional Chinese medicine has reached a several achievements in improving symptoms ,but also plays an irreplaceable role, and it is necessary to summarize and improve the methods of TCM for this disease.Yu-Ping-Feng-Powder (YP), a traditional Chinese medicine prescription, is compo -se d by Radix Astragali, Largehead Atractylodes Rhizome, Divaricate Saposhnikovia Root.It has the effect of invigorating qi and consolidation of exterior, strengthening body resistance and arresting sweating, eliminating pathogen and defending wing pathogen. It is widely applied in China for the cure and prevention of recurrent respiratory tract infections, bronchial asthma (including variant asthma, exercise-induc- ed asthma, asthma in remission stage), allergic cough, bronchitis and other respiratory diseases.The fractioned polysaccharides (YPF-P) isolated from Yu-Ping-Feng-Powder (YP) have immunomodulating activities and that the polysaccharides constitute one of the active components of YP. The studys show YPF-P has the effects of regulation of immune function, anti-bacterial adhesion, the role of inhibition of influenza virus, etc. To further explore the effect of fractioned polysaccharides (YPF-P) isolated from YP on pulmonary fibrosis, the pulmonary fibrosis model was induced by Bleomycin A5 Hydrochloride in Wistar rats. The mechanisms of action of YPF-P on pulmonary fibrosis by using the methods in vivo and molecular levels were studied.The main contents were divided into some sections as follows:1. Intervention effect of YPF-P on pulmonary fibrosis in ratsA total of 90 Wistar rats were randomly divided into the normal control group, the Bleomycin A5 model group, YPF-P 60mg/kg group, YPF-P120mg/kg group and prednisone group. Rats in the model group, YPF-P 60mg/kg group, YPF-P 120mg/kg group and prednisone group were injected with a single dose of Bleomycin A5 by trachea, and rats in the normal control group with the same normal saline. On the 7, 14, 28 days, 6 rats per group were randomly killed. Compared with model group, YPF-P increased weight and inhibited the elevation of index of lung. Furthermore, YPF-P treatment significantly reduced Col-Ⅲ, Col-Ⅳ, HA, LN content in serum and Hyp in lung tissue. Lung histopathological examination showed YPF-P could also ameliorate fibrogenesis and pseudolobule formation by hematoxylin-eosin (HE) staining and masson staining. These results suggest that TFB has positively preventive effects and treatment effects on immunological liver fibrosis by pig serum.2. The mechanisms of Intervention effect of YPF-P on pulmonary fibrosis in ratsFurther study showed that YPF-P treatment markedly reduced MDA in Lung tissue, increased SOD activity. YPF-P treatment decreased TGF-β1 in lung by ELISA. And then YPF-P treatment could significantly down-regulated TGF-β1 and Smads3 mRNA expression and up-regulated Smads7 mRNA expression in lung by RT-PCR analysis.Conclusion: YPF-P significantly had intervention effect on pulmonary fibrosis in rats induced by Bleomycin A5 Hydrochloride probably by its free-radical scavenging ability, inhibition of TGF-β1/Smads pathway and collagen-Ⅰproduction.
