Objective: To investigate the Effects of lignanoids from the acetoacetate extract of Vitex negundo seed (Comp 6,Comp 7,Comp 8 and Comp 10)on the growth and apoptosis of human cervical cancer Hela cells and its mechanism.Methods: Human cervical cancer Hela cell line was cultured in vitro. Nucleic acid synthesis of Hela cells were determined by the incorporation of BrdU. The plate colony formation and the colony formation in soft agar methods wer used to detect the growth of Hela cells. The apoptosis and cell cycle were analyzed by flow cytometry with PI staining. The expression of Mcl-1 protein in Hela cells was observed using indirect immune fluorescence technique with flow cytometry.Results: Four species of lignanoids from the acetoacetate extract of Vitex negundo seed (Comp 6,Comp 7,Comp 8 and Comp 10) all inhibited nucleic acid synthesis of Hela cells, in a concentration-dependent manner. Comp 8 is the strongest of those. IC50 value of that is 0.93μg/mL, and the potency is dimidius of taxinol(Tax) . Comp 6,Comp 7,Comp 8 and Comp 10 all suppressed the growth and colony formation of Hela cells . IC50 of Comp 8 is 0.13μg/mL and 0.57μg/mL, respectively. Four species of lignanoids from the acetoacetate extract of Vitex negundo seed efficiently induced apoptosis and cell cycle arrest in G1/G0 phase of Hela cells. EC50 for induction of apoptosis of Comp 8 is 22.9μg/mL, and similar to Tax(31.0μg/mL). Comp 8 markedly downregulated the expression of Mcl-1 protein in Hela cells, in a concentration-dependent manner. Conclusion:1. The lignanoids from the acetoacetate extract of Vitex negundo seed inhibites nucleic acid synthesis and growth of Hela cells.2. The lignanoids from the acetoacetate extract of Vitex negundo seed can efficiently induce apoptosi of Hela cells.3. The growth inhibitory and apoptosis induced effects of The lignanoids from the acetoacetate extract of Vitex negundo seed in Hela cell line may be involved in the dowmregulation of Mcl-1 protein expression.
