Background:Studies indicate that the following factors may be asso- ciated with atherosclerosis:blood glucose uncontrol,dyslipidemia and inflammation. Abu-lebden et al observed 449 diabetics,and concluded that fasting blood glucose(FBG) is a independent risk facor of coronary artery disease.Some reserch finded that the risk of atherosclerosis increases 11% when glycosylated hemoglobin A1(HBA1C) increases 1%.Diabetics are not only mean hyperglycaemia,most of them might be dyslipidemia.Dys- lipidemia is a critical factor of atherosclerosis.As a new marker of inflammation which link with cardiovascular disease,lipoprotein-associated phosphlipase A2(Lp-PLA2) is being concerned more and more.Gene and environment can both effect plasma Lp-PLA2,and the most effect is a missense mutation (G994T) in exon 9.Recently,some researchers reported that a single point mutation (G994T) in exon 9 of Lp-PLA2 may be a pivotal factor of atherosclerosis in Japanese. Therefore,we observed blood glu- cose,lipid,and missense mutation (G994T) in exon 9 of Lp-PLA2, and explore the possible risk facors of atherosclerosis in 230 diabetics.Objective:To study the possible risk facors of atherosclerosis in Dalian people.Methods:A case-control study including 107 patients with atherosc- lerosis(plaque was detected by Color Doppler Ultrasound),61 men and 46 women which average age is 59.82+10.83 years and 123 controls(no plaque was detected by Color Doppler Ultrasound),67 men and 56 women which average age is 55.40+10.83 years.These subjects selected are excepted for such problems as infection,tumor,sevier liver and renal dysfunction,cardia insufficiency and the prensence of stress factors.All of them are Han people of Dalian in Endocrine wards of the secend affiliated hospital of Dalian Medical University.Compare with age,body mass index (BMI),fasting blood glucose (FBG),glycosylated hemoglobin A1(HBA1C),triglyceride (TG),total cholesterol (TC),high density lipoprotein cholesterol (HDL-C),low density lipoprotein cholesterol (LDL-C),ApolipoproteinA1 (ApoA1),Apolipopro- teinB(ApoB) between the two groups.Simultaneously,extracted peri- pheral blood white blood cell genomic DNA to detect Lp-PLA2 gene mutation by the application of polymerase chain reaction(PCR),and calculate gene- type,allele frequency and difference of each group.Results:In atherosclerosis group FBG, HBA1C and LDL-C were higher than control group.There was significant difference between the two groups.Among controls,the frequencies for the GG,GT and TT were 70.73%,20.27% and 0%,respectively,the frequencies for the G and the T allele were 85.4% and 14.6%.Among patients with atherosclerosis the frequencies for the GG,GT and TT were 57.01%,40.19% and 2.8% ,respectively,the frequencies for the G and the T allele were 77.1% and 22.9%.There was significant difference between the two groups.In atherosclerosis group,T allele frequency was higher than control group.Conclusions:FBG, HBA1C,LDL-C,and a single point mutation (G994T) in exon 9 of plasma Lp-PLA2 gene appear to associated with ather- osclerosis.Our findings need to be confirmed in a larger,prospective study including patients from different populations.
